目的分析慢性粒细胞白血病 ( CML)病人的克隆性增殖 T细胞及其 CDR3序列的特点。方法利用 RT- PCR扩增 3例 CML 外周血单个核细胞中 2 4个 TCR Vβ基因的 CDR3,了解 TCR VβT细胞的分布。阳性产物进一步经基因扫描分析 ,了解其克隆性。寡克隆的 PCR产物再进行序列分析。结果 3例病人仅存在 3~ 9个 TCR Vβ亚家族 T细胞 ,部分 Vβ亚家族T细胞呈寡克隆性增殖 ,Vβ2 1的寡克隆 T细胞见于全部的 3例病人中。结论 CML 病人外周血 TCR Vβ亚家族 T细胞出现倾斜性分布和克隆性增殖 ,这可能是 CML 中机体对恶性细胞所引起的特异性免疫反应 ,但对不同病人的 Vβ2 1寡克隆 T细胞序列分析并未能发现完全同源的 CDR3序列
Objective To analyze the characteristics of clonal proliferating T cells and their CDR3 sequences in patients with chronic myeloid leukemia (CML). Methods The CDR3 of 24 TCR Vβ genes in 3 peripheral blood mononuclear cells of CML were amplified by RT-PCR to understand the distribution of TCR Vβ T cells. The positive product was further analyzed by gene scanning to understand its clonality. Oligoclonal PCR products were subjected to sequence analysis. Results There were only 3 to 9 TCR Vβ subfamily T cells in 3 patients. Some Vβ subfamily T cells showed oligoclonal proliferation. The oligoclonal T cells of Vβ2 1 were found in all 3 patients. Conclusions TCR Vβ subfamily T cells in peripheral blood of patients with CML have tilted distribution and clonal proliferation, which may be the specific immune response induced by the malignant cells in CML. However, sequence analysis of Vβ21 oligoclonal T cells in different patients And failed to find completely homologous CDR3 sequences
