目的:通过观察苏木乙酸乙酯提取物对同种异位心脏移植急性排斥反应模型大鼠移植凋亡相关蛋白Bcl-2/Bax表达的影响,探讨其免疫抑制作用的机理,为苏木的新药研发提供重要的理论和实验依据。方法:以Wistar大鼠为供体,SD大鼠为受体,制备大鼠同种异位心脏移植模型,造模成功后,采用免疫组织化法检测Bcl-2/Bax蛋白的表达。结果:与模型对照组比较,各治疗组Bcl-2蛋白的表达未见明显改变,差异无统计学意义。苏木组与模型对照组比较,能明显降低Bax蛋白的表达,二者比较有显著性差异,CsA组和苏木加半量CsA组与模型对照组比较,降低Bax蛋白的表达作用更强,有显著性差异;苏木组比CsA组作用弱,二者比较有显著性差异,苏木加半量CsA组比CsA组作用强,但二者比较无显著性差异。结论:在大鼠心脏移植模型中,苏木乙酸乙酯提取物能下调Bax蛋白的表达,从而发挥抑制心肌细胞凋亡的作用。
OBJECTIVE: To observe the effect of ethyl acetate extract on the expression of apoptosis related protein Bcl-2 / Bax in acute rejection rat model of allograft heart transplantation and to investigate its mechanism of immunosuppression, Research and development of new drugs provide important theoretical and experimental evidence. Methods: Wistar rats were used as donors, SD rats were used as recipients to prepare rat allograft heart transplantation model. After successful modeling, the expression of Bcl-2 / Bax protein was detected by immunohistochemistry. Results: Compared with the model control group, the expression of Bcl-2 protein in each treatment group showed no significant change, the difference was not statistically significant. Compared with the model control group, the expression of Bax protein in Hematoxylin-Eosin group was significantly lower than that in the model control group, and there was significant difference between the two groups (P <0.05). Compared with the model control group, CsA group and Hematoxylin- Significant difference; Hematoxylin than CsA group weaker, the two were significantly different, the hematoxylin and the amount of CsA CsA group stronger than CsA group, but there was no significant difference between the two. Conclusion: In the rat heart transplantation model, ethyl acetate extract of Hematoxylin and Eosin can down-regulate the expression of Bax protein and thus play an inhibitory role in cardiomyocyte apoptosis.
