目的:探讨RNA干扰沉默EZH2基因对人卵巢癌细胞系A2780增殖和迁移的影响。方法:真核表达载体介导靶向沉默EZH2 shRNA的重组质粒转染A2780细胞,Real-time RT-PCR检测EZH2基因的沉默效果,MTT法检测细胞增殖,流式细胞仪检测细胞周期,Transwell小室检测细胞迁移。结果:EZH2shRNA转染明显下调EZH2的表达(P<0.001)。重组质粒稳定转染至A2780细胞后,细胞增殖速度明显下降(P<0.05),G0/G1期细胞增加(P<0.01),体外迁移能力下降(P<0.01),差异均有统计学意义。结论:EZH2基因沉默能有效抑制卵巢癌细胞的增殖和迁移能力,为卵巢癌的表观遗传学治疗提供新靶点。
Objective: To investigate the effect of RNAi silencing EZH2 gene on the proliferation and migration of human ovarian cancer cell line A2780. METHODS: A2780 cells were transfected with recombinant plasmid targeting EZH2 shRNA targeting eukaryotic expression vector. The silencing effect of EZH2 gene was detected by Real-time RT-PCR. Cell proliferation was detected by MTT assay. Cell cycle was detected by flow cytometry. Transwell chamber Cell migration was examined. Results: EZH2shRNA transfection significantly down-regulated the expression of EZH2 (P <0.001). After A2780 cells were stably transfected with the recombinant plasmid, the cell proliferation rate was significantly decreased (P <0.05), G0 / G1 phase cells increased (P <0.01), migration in vitro decreased (P <0.01), the difference was statistically significant. Conclusion: EZH2 gene silencing can effectively inhibit the proliferation and migration of ovarian cancer cells and provide a new target for the epigenetic treatment of ovarian cancer.
