目的明确Ghrelin基因Leu72Met多态性在中国甘肃省东乡族的分布并探讨其与2型糖尿病的关系。方法用PCR-DHPLC技术和测序法检测甘肃省东乡族T2DM患者及正常对照者(NC组)Ghrelin基因Leu72Met多态性。结果 Leu72Met基因型和等位基因频率在T2DM组和NC组中的分布差异无统计学意义(P>0.05);NC组cA+AA型的Fins、HOMA-IR明显高于CC型;T2DM组CC基因型的FPG、血肌酐(SCr)水平明显高于cA+AA型(P均<0.05)。结论 Ghrelin基因Leu72Met多态性可能参与胰岛素抵抗和2型糖尿病的发病机制。
Objective To determine the distribution of Leu72Met polymorphism of Ghrelin gene in Dongxiang ethnic group in Gansu Province of China and explore its relationship with type 2 diabetes mellitus. Methods The Ghrelin gene Leu72Met polymorphism in T2DM patients of Dongxiang nationality in Gansu province and normal controls (NC group) was detected by PCR-DHPLC and sequencing. Results There was no significant difference in genotype and allele frequencies of Leu72Met in T2DM and NC (P> 0.05). Fins and HOMA-IR of cA + AA in NC were significantly higher than those in CC Genotype FPG, serum creatinine (SCr) levels were significantly higher than cA + AA type (P all <0.05). Conclusion Ghrelin gene Leu72Met polymorphism may be involved in the pathogenesis of insulin resistance and type 2 diabetes.
