目的检测再生障碍性贫血(再障)患者单个CD8+T细胞内干扰素-γ(IFN-γ)的绝对含量。方法用毛细管电泳免疫分析激光诱导荧光法对再障患者外周血单个CD8+T细胞内IFN-γ的水平进行定量检测。结果2例重型再障(SAA)患者免疫治疗前外周血单个CD8+T细胞内IFN-γ的水平分别为(151.53±28.92)z mol和(223.72±45.23)z mol,免疫治疗后相应的降为(45.57±13.90)z mol和(36.44±9.66)z mol,正常对照为(47.47±17.97)z mol。经统计分析,SAA患者治疗前、后及与正常对照比较差异均有统计学意义。结论毛细管电泳免疫分析激光诱导荧光法可以检测到外周血单个CD8+T细胞内IFN-γ的z mol水平,有望成为一种新的检测方法应用于临床。
Objective To determine the absolute content of interferon-γ (IFN-γ) in single CD8 + T cells in patients with aplastic anemia (aplastic anemia). Methods The level of IFN-γ in peripheral blood mononuclear cells (CD8 + T cells) in patients with aplastic anemia was quantitatively determined by capillary electrophoresis immunoassay and laser induced fluorescence spectrometry. Results The levels of IFN-γ in peripheral blood CD8 + T cells in peripheral blood of two patients with SAA were (151.53 ± 28.92) z mol and (223.72 ± 45.23) z mol respectively before the immunotherapy. The corresponding reduction after immunotherapy (45.57 ± 13.90) z mol and (36.44 ± 9.66) z mol respectively, and the normal control was (47.47 ± 17.97) z mol. After statistical analysis, SAA patients before and after treatment and compared with the normal control were statistically significant differences. Conclusion Capillary electrophoresis immunoassay laser-induced fluorescence method can detect the z mol level of IFN-γ in peripheral blood CD8 + T cells, which is expected to become a new detection method in clinic.
