目的:建立以高效凝胶过滤色谱法(HPGFC)测定沙漠嘎多糖重均相对分子质量的方法。方法:通过将沙漠嘎多糖分离、柱层析纯化,获得沙漠嘎多糖S1,采用HPGFC法测定重均相对分子质量。色谱条件:Asahipak GS色谱柱GS-3207E(250mm×7.5mmID,9μm)与GS-6207G(500mm×7.5mmID,9μm)串联使用;流动相:0.003mol·L-1乙酸钠;流速:1mL·min-1;柱温:35℃;示差折光检测器(RID)。结果:沙漠嘎多糖重均相对分子质量的回归方程为:lg(Mw)=10.167-0.287tR(r=-0.976),线性范围为:10000~500000Da,测得沙漠嘎多糖S1重均相对分子质量约为8.5×104Da。结论:为沙漠嘎多糖重均相对分子质量测定提供了简便、实用的分析方法。
OBJECTIVE: To establish a method for the determination of weight average molecular weight of desert polysaccharides by high performance gel filtration chromatography (HPGFC). Methods: Desert Ga polysaccharide was isolated and purified by column chromatography to obtain desert Ga polysaccharide S1. The weight average molecular weight was determined by HPGFC method. Chromatographic conditions: Asahipak GS column GS-3207E (250mm × 7.5mmID, 9μm) was used in series with GS-6207G (500mm × 7.5mmID, 9μm); mobile phase was 0.003mol·L-1 sodium acetate; -1; column temperature: 35 ° C .; refractive index detector (RID). Results: The regression equation of weight average relative molecular weight of GA in the desert was: lg (Mw) = 10.167-0.287tR (r = -0.976) and the linear range was 10000-500000Da. The weight average relative molecular mass About 8.5 × 104 Da. Conclusion: It provides a simple and practical method for the determination of weight-average molecular weight of GAPS.
