目的观察Toll样受体4(TLR4)/核转录因子kappa B(NF-κB)信号通路及其相关炎症因子对周围性面神经炎的影响。方法 50例周围性面神经炎患者为试验组,50例同期健康体检者为对照组。用实时荧光定量聚合酶链式反应(RT-q PCR)和蛋白质印迹法(Western blot)检测外周血单核细胞(PBMCs)中TLR4 mRNA及蛋白;Western blot检测PBMCs中NF-κB p65磷酸;酶联免疫吸附法(ELISA)检测血清中肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)。结果试验组和对照组PBMCs中TLR4 mRNA的相对表达水平分别为1.55±0.25,0.83±0.10;TLR4蛋白的相对表达水平分别为1.11±0.17,0.61±0.11;试验组和对照组PBMCs中NF-κB p65磷酸化水平分别为0.67±0.30,0.36±0.22;TNF-α分别为(149.50±12.12),(102.10±10.60)ng·L-1;IL-1β分别(255.50±19.19),(105.20±12.80)ng·L~(-1),差异均有统计学意义(P<0.05)。结论 PBMCs中TLR4/NF-κB信号通路的活化及相关炎症因子的表达变化可能在周围性面神经炎的发生发展中发挥作用。
Objective To observe the effects of Toll-like receptor 4 (TLR4) / nuclear factor kappa B (NF-κB) signaling pathway and its related inflammatory cytokines on peripheral facial neuritis. Methods 50 cases of peripheral facial neuritis patients for the experimental group, 50 cases of the same period of health examination for the control group. TLR4 mRNA and protein in peripheral blood mononuclear cells (PBMCs) were detected by real-time fluorescence quantitative polymerase chain reaction (RT-q PCR) and Western blot; Western blot was used to detect NF-κB p65 phosphorylation in PBMCs; Serum levels of tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) were detected by ELISA. Results The relative expression levels of TLR4 mRNA in PBMCs of experimental group and control group were 1.55 ± 0.25 and 0.83 ± 0.10, respectively. The relative expression levels of TLR4 protein were 1.11 ± 0.17 and 0.61 ± 0.11, respectively. NF-κB The levels of p65 phosphorylation were 0.67 ± 0.30 and 0.36 ± 0.22, respectively. The levels of TNF-α were (149.50 ± 12.12) and (102.10 ± 10.60) ng · L -1, respectively. The levels of IL-1β were 255.50 ± 19.19 and 105.20 ± 12.80 ) ng · L ~ (-1), the differences were statistically significant (P <0.05). Conclusion The activation of TLR4 / NF-κB signaling pathway and the expression of related inflammatory factors in PBMCs may play a role in the occurrence and development of peripheral facial neuritis.
