目的 :研究缬沙坦对自发性高血压大鼠 (SHR)左室心肌整合素 β1和纤维黏连蛋白 (FN)表达的影响 ,探讨高血压心肌纤维化的机制。方法 :选用 6周龄的雄性SHR 12只 ,随机均分为 2组 :SHR阳性对照组 ;缬沙坦干预组 (SHR V) ,灌喂缬沙坦 2 0mg·kg-1·d-1。另选同源正常血压的WKY大鼠 6只为正常对照组。实验期 14周。观察血压、左室重量 /体重 (LVW /BW )、胶原容积分数 ,并用免疫组化法检测 3组大鼠心肌整合素 β1和FN的表达。结果 :缬沙坦可显著降低SHR的血压、LVW /BW及左室心肌的胶原含量 ,此外SHR阳性对照组整合素 β1和FN的表达明显高于正常对照组 (P <0 .0 5 ) ,而SHR V组两者的表达显著低于SHR阳性对照组 (P <0 .0 5 )。结论 :缬沙坦除可降低血压外 ,还能显著抑制整合素 β1和FN的表达 ,抑制甚至逆转心肌纤维化过程
Objective: To investigate the effects of valsartan on the expression of integrin β1 and fibronectin (FN) in left ventricular myocardium in spontaneously hypertensive rats (SHR) and to explore the mechanism of myocardial fibrosis in hypertensive rats. Methods: Twelve male SHRs of 6 weeks old were randomly divided into 2 groups: SHR-positive control group, valsartan intervention group (SHR V) and valsartan 20 mg · kg-1 · d-1. Another 6 normal WKY rats with normal blood pressure were selected as normal control group. The experimental period of 14 weeks. The blood pressure, left ventricular mass / body weight (LVW / BW) and collagen volume fraction were observed. The expression of myocardial integrin β1 and FN were detected by immunohistochemistry. Results: Valsartan significantly decreased the blood pressure, LVW / BW and the collagen content of left ventricular myocardium in SHR group. In addition, the expression of integrin β1 and FN in SHR positive control group was significantly higher than that in normal control group (P <0.05) However, the expression of SHR V in both groups was significantly lower than that in SHR positive control group (P <0.05). Conclusion: Valsartan can not only reduce blood pressure, but also significantly inhibit the expression of integrin β1 and FN, and inhibit or even reverse the process of myocardial fibrosis
