目的:观察消癥散对人肝癌HepG-2细胞凋亡的影响,探讨其抗肿瘤的可能作用机制。方法:MTT比色法检测消癥散对人肝癌HepG-2细胞的生长抑制作用;倒置显微镜观察细胞形态学的改变;RT-PCR法检测消癥散对人肝癌HepG-2细胞p53和Bcl-2 mRNA表达的影响。结果:消癥散对人肝癌细胞HepG-2的生长活性有抑制作用,并可调节人肝癌HepG-2细胞中p53和Bcl-2 mRNA的表达水平。结论:消癥散能促进人肝癌HepG-2细胞凋亡,其作用机制可能与下调Bcl-2和升高p53相关。
Objective: To observe the effect of Xiaozheng San on the apoptosis of human hepatocellular carcinoma HepG-2 cells and to explore the possible mechanism of anti-tumor effect. Methods MTT assay was used to detect the growth inhibitory effect of Xiaozheng San on human hepatocellular carcinoma HepG-2 cells. The morphological changes of cells were observed by inverted microscope. The effects of Xiaozheng San on p53 and Bcl- 2 mRNA expression. Results: Xiaozisan could inhibit the growth of HepG-2 cells and regulate the expression of p53 and Bcl-2 mRNA in HepG-2 cells. Conclusion: Xiaoshi Powder can promote the apoptosis of human hepatocellular carcinoma HepG-2 cells. The mechanism may be related to the down-regulation of Bcl-2 and up-regulation of p53.
