目的检测熊果酸对骨肉瘤细胞生长转移的影响。方法将不同浓度熊果酸分别作用于人骨肉瘤细胞系U2OS细胞,MTT实验检测细胞增殖情况,Hochest 33258染色检测细胞凋亡情况,Transwell实验检测细胞转移情况。利用Western blot方法检测熊果酸对相关蛋白的作用。结果 10、20、30μg/mL熊果酸处理U2OS细胞24 h后,对其增殖的抑制率分别为39.51%±0.75%、48.91%±3.64%、56.49%±1.54%。熊果酸通过抑制cyclin d1蛋白抑制U2OS细胞的增殖,通过调节bcl-2和bax蛋白促进U2OS细胞的凋亡。Transwell实验发现,熊果酸可以剂量依赖的方式抑制U2OS细胞的迁移,其抑制作用是通过其对MMP2蛋白的抑制实现的。结论熊果酸可以抑制骨肉瘤细胞系U2OS细胞的增殖与转移,可以作为临床骨肉瘤治疗的潜在化疗药物进行进一步探索。
Objective To detect the effect of ursolic acid on the growth and metastasis of osteosarcoma cells. Methods Different concentrations of ursolic acid were applied to human osteosarcoma cell line U2OS cells respectively. MTT assay was used to detect cell proliferation. Hochest 33258 staining was used to detect cell apoptosis. Transwell assay was used to detect the cell metastasis. The effect of ursolic acid on related proteins was detected by Western blot. Results U2OS cells treated with 10, 20 and 30 μg / mL ursolic acid for 24 h showed inhibitory rates of 39.51% ± 0.75%, 48.91% ± 3.64% and 56.49% ± 1.54%, respectively. Ursolic acid inhibits the proliferation of U2OS cells by inhibiting the cyclin d1 protein and promotes the apoptosis of U2OS cells by regulating the bcl-2 and bax proteins. Transwell experiments found that ursolic acid inhibits the migration of U2OS cells in a dose-dependent manner by its inhibition of MMP2 protein. Conclusion UA can inhibit the proliferation and metastasis of the osteosarcoma cell line U2OS and can be further explored as a potential chemotherapeutic for the treatment of osteosarcoma.
