淡水养殖鱼类结节病是近年来时常爆发的疾病之一,为了有效预防和防治该病,从合肥郊区某养殖场濒死的黄金鲫(鲤鱼♀×鲫鱼♂)体内分离优势致病菌(命名为AH菌株),对其进行了鉴定。采用传统细菌分类方法,测定该菌株的形态特征、生理生化特性和常见药物的敏感性;应用分子生物学方法,对病原菌16S rRNA特异片段进行克隆、序列测定分析。AH菌株与对照鮰爱德华氏菌在形态特征和生理生化特性基本一致。序列分析结果显示分离的AH菌株与鮰爱德华氏菌(AB050826,NR_024769)菌株间基因序列同源性为99.9%以上,在系统发育树上与鮰爱德华氏菌聚为一族。人工回归复感染健康黄金鲫鱼,出现与原发性相似的内脏类结节病症状。药敏实验结果显示:AH菌株对氧氟沙星等11种药物敏感,对四环素等4种抗生素耐药,对磺胺类药物复方新诺明中度敏感。综合分析认为鮰爱德华氏菌是黄金鲫内脏类结节病的重要致病菌,该结果为黄金鲫内脏类结节病有效防控提供有益参考。
Freshwater cultured fish sarcoidosis is one of the frequently erupted diseases in recent years. In order to effectively prevent and cure this disease, the predominant pathogenic bacteria (carp ♀ × crucian carp ♂) were isolated from the moribund gold crucian carp Named AH strain), its identification. The traditional bacterial classification method was used to determine the morphological characteristics, physiological and biochemical characteristics of the strain and the sensitivity of common drugs. Clone 16S rRNA specific fragment of pathogenic bacteria by molecular biology method and sequence analysis. AH strain and control 鮰 Edwardsiella in morphological characteristics and physiological and biochemical characteristics of the same. The results of sequence analysis showed that the homology between the isolated AH strain and E. bovis (AB050826, NR_024769) was above 99.9%, and the phylogenetic tree was clustered with R. ederiva. Artificial regression complex infection of healthy gold carp, appeared similar to the primary visceral nodular disease symptoms. The results of drug sensitivity test showed that AH strain was sensitive to 11 kinds of drugs such as ofloxacin, resistant to 4 kinds of antibiotics such as tetracycline and moderately to sulfamethoxazole. Comprehensive analysis shows that E. bovis is an important causative agent of visceral sarcoidosis of golden crucian carp. The results provide a useful reference for the effective prevention and control of visceral sarcoidosis of golden crucian carp.
