目的探讨牡荆苷对体外培养大鼠骨髓间充质干细胞成骨分化的影响。方法采用MTT法检测牡荆苷对骨髓间充质干细胞生存率的影响,以对硝基苯磷酸盐法检测碱性磷酸酶活性,茜素红法染色评价钙化结节形成。实时荧光定量PCR检测runt-related transcription factor 2(Runx2)和osterix(Osx)表达。结果牡荆苷在0.1~50μmol·L?1浓度内对骨髓间充质干细胞生存率无显著影响。牡荆苷能明显提高骨髓间充质干细胞内碱性磷酸酶活性,并能促进钙化结节形成。牡荆苷也上调Runx2和Osx基因表达。结论牡荆苷可促进骨髓间充质干细胞向成骨细胞方向分化,其作用可能与上调Runx2和Osx基因表达有关。
Objective To investigate the effect of vitexin on osteogenic differentiation of rat bone marrow mesenchymal stem cells in vitro. Methods MTT assay was used to detect the effect of vitexin on the survival rate of bone marrow mesenchymal stem cells. The activity of alkaline phosphatase was assayed by p-nitrophenyl phosphate method and the calcified nodules were evaluated by alizarin red staining. Real-time fluorescence quantitative PCR was used to detect the expression of runt-related transcription factor 2 (Runx2) and osterix (Osx). Results Vitexin had no significant effect on the survival rate of bone marrow mesenchymal stem cells in the concentration of 0.1 ~ 50μmol·L -1. Vitexin can significantly improve alkaline phosphatase activity in bone marrow mesenchymal stem cells, and can promote the formation of calcified nodules. Vitexin also up-regulated Runx2 and Osx gene expression. Conclusion Vitexin can promote the differentiation of bone marrow mesenchymal stem cells into osteoblasts, which may be related to the up-regulation of Runx2 and Osx gene expression.
