目的:探讨年龄对大鼠原代培养的阴茎海绵体平滑肌细胞(CCSMCs)分泌胰岛素样生长因子-1(IGF-1)水平的影响,及其与老年性ED的关系。方法:体外原代培养不同年龄段(4月龄、12月龄和24月龄)的大鼠CCSMCs,免疫组化染色鉴定;定量培养CCSMCs,采用酶免疫分析法(EIA)定量检测CCSMCs分泌IGF-1含量的变化,分析比较不同年龄对CCSMCs分泌IGF-1含量的影响。结果:成功原代培养CCSMCs,随着月龄的增加,CCSMCs分泌的IGF-1含量减少,与4月龄(7.1 ng/105个细胞)比较、12月龄(2.2 ng/105个细胞)和24月龄(1.9ng/105个细胞)CCSMCs分泌的IGF-1含量明显减少,有统计学意义(P<0.01)。结论:年龄是影响原代培养CCSMCs分泌IGF-1水平的因素之一,IGF-1的低表达可能和老年性ED的发生有关。
AIM: To investigate the effect of aging on the level of insulin-like growth factor-1 (IGF-1) secreted by primary cultured rat corpus cavernosum smooth muscle cells (CCSMCs) and its relationship with senile ED. Methods: Primary cultured rat CCSMCs of different ages (4 months, 12 months and 24 months old) were identified by immunohistochemical staining. CCSMCs were cultured quantitatively and quantitatively detected by enzyme immunoassay (EIA) -1 content of CCSMCs were analyzed. The effects of different ages on the secretion of IGF-1 in CCSMCs were compared. Results: After primary culture of CCSMCs, the IGF-1 secretion of CCSMCs decreased with the increase of age. Compared with 4-month old (7.1 ng / 105 cells), 12-month old The levels of IGF-1 secreted by CCSMCs at 24 months (1.9ng / 105 cells) were significantly decreased (P <0.01). Conclusion: Age is one of the factors that affect the secretion of IGF-1 in primary cultured CCSMCs. The low expression of IGF-1 may be related to the occurrence of ED in senile patients.
