目的 了解北京市同性恋HIV-1感染者HIV-1的亚型类型及传播来源和流行时间。方法 应用套式聚合酶链式反应(PCR)对12份1993~2001年北京市HIV-1阳性同性恋者外周血单个核细胞(PBMC)的核酸样品进行扩增,并对其包膜区的C2-V3段的306个核酸序列进行测定和分析。结果12份样品全部是B亚型的HIV-1毒株序列,其亚型内的基因离散率为 10.35± 2.06,与国际 A-E亚型共享序列比较后发现其与 A、C、D、E亚型的共享序列的基因离散率均大于 25%,而与国际 B亚型共享序列的基因离散率仅为11.25±3.60。系统树分析显示,12个毒株与B亚型共享序列聚在一起并远离其它国际亚型,并且12个毒株与SF162紧密相连,而与国际B亚型共享序列和泰国B亚型代表株TH14可以分开。对gp120中最重要的中和抗体决定簇V3环序列进行对比分析发现,12毒株在V3环中变化较大,其中4毒株带有GPGR这一欧美B亚型V3环顶端四肽序列特征,占33.33%,1个毒株带有GLGR,占8.33%,而其它7个毒株为GWGR,占58.34%。结论HIV-1在北京市同性恋人群中流行的为B亚型,流行来源为欧美,流行时间10年左右,V3环顶端四肽序列特征以GWGR为主。
Objective To understand the subtypes, sources and prevalence of HIV-1 in homosexual HIV-1 infected persons in Beijing. Methods Nucleotide samples of 12 peripheral blood mononuclear cells (PBMCs) from HIV-1 positive homosexuals in Beijing from 1993 to 2001 were amplified by nested polymerase chain reaction (PCR) and their C2 The 306 nucleic acid sequences of the -V3 segment were assayed and analyzed. Results All the 12 samples were subtype B of HIV-1 strain. The subgenotype of the HIV-1 strain was 10.35 ± 2.06. Compared with the shared sequence of the subtype AE, Type gene shared more than 25% of the gene share, while the gene share with the international subtype B only had a genetic divergence rate of only 11.25 ± 3.60. Phylogenetic tree analysis showed that 12 strains were clustered together with subtype B and distant from other international subtypes, and 12 strains were closely linked with SF162. However, shared sequences with international subtype B and representative strains with subtype B TH14 can be separated. Comparative analysis of the V3 loop sequences of the most important neutralizing antibody determinants in gp120 revealed that the 12 strains varied greatly in the V3 loop, of which 4 strains had the GPGR characteristic of the V3 loop top tetrapeptide in European and American B subtypes , Accounting for 33.33%. One strain had GLGR accounting for 8.33%, while the other seven strains were GWGR accounting for 58.34%. Conclusions HIV-1 is endemic to the same-sex population in Beijing. The epidemic source is Europe and the United States. The prevalence of HIV-1 is about 10 years. The top tetrapeptide sequence of V3 loop is characterized by GWGR.
