通过对新疆紫草Arnebia euchroma红白2种色系悬浮细胞的表型、生长曲线、次生代谢产物积累研究,用数学建模模拟新疆悬浮细胞的增殖和紫草素类化合物积累情况,研究新疆紫草悬浮细胞生长和次生代谢生产的动力学参数,发现红色高产系为针对紫草素类次生代谢产物生产的优良细胞株系,白色低产系可能为突变株系。用Logistic模型对生长函数进行拟合并求导数,得到新疆紫草悬浮细胞的生长速率及生长加速度变化曲线,确定了新疆紫草悬浮细胞继代培养最佳时期为继代到新鲜培养基中的第15天,给诱导处理的最佳时期为继代到新鲜培养基中后第13~14天。并且通过比较新疆紫草红色系细胞增长速率的变化趋势与紫草素类化合物积累曲线发现,新疆紫草悬浮7细胞红色系生物量快速增长时不利于紫草素的合成与积累。
Through the research on the phenotype, growth curve and secondary metabolite accumulation of Arnebia euchroma red-and-white color suspension cells in Xinjiang, we studied the proliferation of Xinjiang suspension cells and the accumulation of shikonin by mathematical modeling. Lithospermum cell growth and secondary metabolism of kinetic parameters of the production and found that the red high yield line for shikonin secondary metabolites production of excellent cell lines, white low-yield lines may be mutant strains. The Logistic model was used to fit the growth function and derive the derivative. The growth curve and the acceleration curve of the growth of Lithospermum viridis were obtained, and the best subculture of Lithospermum abalone was identified as the subculture medium On the 15th day, the optimal period for inducing treatment was from the 13th to the 14th day after subculture to the fresh medium. And by comparing the trend of the growth rate and the shikonin accumulation curve of Lithospermum erythrocytic red line in Xinjiang, it was found that the rapid growth of Lithospermum line 7 suspension cells was not conducive to the synthesis and accumulation of Shikonin.
