目的:研究灵杞黄斑颗粒对大鼠光损伤模型视网膜细胞凋亡相关基因c-fos、caspase-3及bcl-xL表达的影响。方法:SD大鼠随机分为正常对照组、光损伤给水组、光损伤给药组。采用白色荧光灯持续照射5h建立光损伤模型,于光照2·5h、5h及光照后1d及3d,通过逆转录-聚合酶链反应(RT-PCR)和免疫蛋白印迹(western blot)分析视网膜c-fos、caspase-3及bcl-xL的转录表达。结果:c-fos、caspase-3mRNA和蛋白表达下调,bcl-xL mRNA和蛋白表达上调。结论:灵杞黄斑颗粒可调控光损伤大鼠视网膜细胞凋亡相关基因的表达。
Objective: To study the effect of Lingqi Huangkuang Granule on the expression of c-fos, caspase-3 and bcl-xL in rat retina after photodamage. Methods: SD rats were randomly divided into normal control group, light injury water group and light injury group. The model of photodamage was established by continuous illumination of white fluorescent lamp for 5h. The changes of c-actin in retinal cortex were detected by reverse transcription-polymerase chain reaction (RT-PCR) and western blot at 2.5h, 5h and 1d, fos, caspase-3 and bcl-xL transcriptional expression. Results: The expressions of c-fos, caspase-3 mRNA and protein were down-regulated while the expressions of bcl-xL mRNA and protein were up-regulated. Conclusion: Lingqi macular particles can regulate the expression of retinal apoptosis-related genes in photodamaged rats.
