目的:探讨药用爬山虎对牛II型胶原诱导性关节炎大鼠(CIA)关节滑膜细胞核周因子-κB(NF-κB)表达及血清IL-17水平的影响。方法:将40只6周龄雌性wistar大鼠随机分为空白对照组、模型对照组、雷公藤多苷对照组和爬山虎治疗组各10只。除空白对照组外,其余各组建立CIA模型,造模第7天开始予爬山虎汤剂和雷公藤水溶液灌胃治疗,连续用药4周,空白对照组和模型对照组灌服等量生理盐水,于末次给药2h后采集血清,处死大鼠,摘取双后肢膝关节分离滑膜,采用ELISA检测血清IL-17的表达,Western-blot检测关节滑膜NF-κB/P65的蛋白表达。结果:与空白对照组比较,模型组大鼠血清IL-17表达增加,爬山虎和雷公藤能够明显降低IL-17的表达(P<0.05);模型对照组大鼠关节滑膜细胞NF-κB/P65蛋白水平明显高于空白对照组(P<0.05),爬山虎与雷公藤多苷治疗组均能降低CIA大鼠关节滑膜细胞NF-κB/P65的表达,两组无显著性差异(P>0.05)。结论:药用爬山虎能够降低CIA模型大鼠血清IL-17的表达,下调关节滑膜细胞NF-κB的蛋白水平。
Objective: To investigate the effects of medicinal ivy on the expression of NF-κB and the level of serum IL-17 in the synoviocytes of CIA rats. Methods: Forty female wistar rats of 6 weeks old were randomly divided into blank control group, model control group, tripterygium glycosides control group and ivy group. Except for the blank control group, CIA model was established in the remaining groups. On the 7th day after modeling, ivy and Tripterygium wilfordii were intragastrically administrated for 4 weeks. The blank control group and the model control group were given the same amount of normal saline, The serum was collected 2 h after the last administration, and the rats were sacrificed. The synovium of both hindlimbs was isolated and the serum IL-17 was detected by ELISA. The protein expression of NF-κB / P65 was detected by Western-blot. Results: Compared with the blank control group, the expression of IL-17 was increased in the model group and ivy and tripterygium wilfordii significantly decreased the expression of IL-17 (P <0.05). The NF-κB / P65 protein levels were significantly higher than the blank control group (P <0.05), ivy and tripterygium glycosides treatment group can reduce the expression of NF-κB / P65 in synovial cells of CIA rats, there was no significant difference between the two groups (P> 0.05). Conclusion: Medicinal ivy can reduce the serum level of IL-17 in CIA model rats and down-regulate the protein level of NF-κB in synovial cells.
