目的:建立用高效液相-库仑阵列电化学检测器测定唾液中对乙酰氨基酚浓度的方法。方法:受试者服用对乙酰氨基酚片,分别于不同时间点取受试者唾液经处理,以甲醇(加5%的水相)与水相(0.03mol.L-1磷酸二氢钠溶液,氢氧化钠溶液和磷酸溶液调pH至6.0)(20:80)为流动相,流速1mL.min-1,350mV电压下检测。结果:对乙酰氨基酚在8.3~833.3ng.mL-1范围内线性关系良好,最低检测限为3.3ng.mL-1,平均回收率、重复性等均符合方法学要求。结论:高效液相-库仑阵列电化学法操作简便、准确,灵敏度高,适用于对乙酰氨基酚体内代谢研究。
Objective: To establish a method for the determination of paracetamol in saliva by HPLC-coulometric array electrochemical detector. Methods: The subjects took paracetamol tablets, respectively, at different time points to take the subjects saliva treated with methanol (plus 5% aqueous phase) and the aqueous phase (0.03mol. L-1 sodium dihydrogen phosphate solution , Sodium hydroxide solution and phosphoric acid solution to adjust pH to 6.0) (20:80) as the mobile phase, the flow rate of 1mL.min-1,350mV voltage detection. Results: Acetaminophen had a good linearity in the range of 8.3-833.3ng.mL-1 with the minimum detectable limit of 3.3ng.mL-1. The average recovery and repeatability all met the methodological requirements. Conclusion: HPLC - Coulomb array electrochemical method is simple, accurate and sensitive, suitable for the study of acetaminophen metabolism in vivo.
