目的探讨灯盏花素对糖尿病大鼠肾脏细胞凋亡及对bax和bcl-2mRNA表达的影响。方法健康雄性SD大鼠随机分为正常对照组(NC)、糖尿病组(DM)和灯盏花治疗组(EB),EB组行灯盏花素20mg·kg-1·d-1腹腔内注射。给药后2周和6周处死,检测肾脏细胞凋亡、肾脏bax和bcl-2mRNA的表达。结果与NC组比较,(1)DM组及EB组肾小管凋亡细胞数明显增多,且DM组高于EB组;(2)DM组肾脏bax、bcl-2mRNA的表达显著增强,bax/bcl-2值明显升高;EB组与DM组比较bcl-2mRNA表达增强,baxmRNA减弱,bax/bcl-2比值降低;(3)EB组及DM组体重下降,肾指数、肾小球面积、体积升高,且EB组较DM组体重增加,肾指数、肾小球面积、体积下降。血尿素氮、肌酐比较:DM组>EB组>NC组。结论灯盏花素可能通过影响凋亡相关基因bcl-2和bax的表达而抑制肾脏细胞凋亡,从而发挥肾脏保护作用。
Objective To investigate the effects of erigeron breviscapus on renal cell apoptosis and the expression of bax and bcl-2 mRNA in diabetic rats. Methods Healthy male Sprague-Dawley rats were randomly divided into normal control group (NC), diabetic group (DM) and Erigeron breviscapus treatment group (EB). EB group was injected with breviscapine 20 mg·kg-1·d-1 intraperitoneally. At 2 and 6 weeks after dosing, they were sacrificed and renal cell apoptosis, kidney bax and bcl-2 mRNA expression were measured. Results Compared with the NC group, (1) the number of apoptotic cells in the renal tubules of the DM group and the EB group increased significantly, and the DM group was higher than the EB group; (2) The expression of bax and bcl-2 mRNA was significantly increased in the DM group, bax/bcl -2 value increased significantly; EB group compared with DM group bcl-2 mRNA expression increased, baxmRNA weakened, bax/bcl-2 ratio decreased; (3) EB group and DM group weight loss, kidney index, glomerular area, volume Elevated, and EB group compared with DM group weight gain, kidney index, glomerular area, volume decreased. Blood urea nitrogen, creatinine comparison: DM group> EB group> NC group. Conclusion Breviscapine may inhibit the apoptosis of renal cells by affecting the expression of apoptosis-related genes bcl-2 and bax, thus exerting renal protective effects.
