目的 :研究uPA系统 4种主要成分 (uPA、PAI 1、PAI 2、uPAR)在肺癌细胞裸鼠移植瘤侵袭转移过程中的作用。方法 :将 4株肺癌细胞 (1株肺巨细胞癌、2株肺腺癌、1株肺鳞癌 )接种入裸鼠体内 ,建立体内侵袭转移模型 ;应用免疫组化、蛋白免疫印迹 (WesternBlot)等方法检测移植瘤组织中uPA系统各因子的表达。结果 :肺巨细胞癌细胞所形成移植瘤的侵袭转移能力最强 ;免疫组化和蛋白免疫印迹 (WesternBlot)均表明uPA系统各因子在 4株移植瘤肿瘤细胞的胞浆及胞膜均有相当的表达 ,在肺巨细胞癌细胞所形成移植瘤中 4种因子的表达水平相对较低 ,而在两株腺癌移植瘤中相对较高。结论 :uPA系统内 4种因子在移植瘤中以多种复合物及单体的形式存在 ;这些因子的表达水平不能确切反映各移植瘤在裸鼠体内侵袭转移能力的差异
Objective: To study the effects of uPA, PAI 1, PAI 2 and uPAR on the invasion and metastasis of lung cancer in nude mice. METHODS: Four lung cancer cells (1 lung giant cell carcinoma, 2 lung adenocarcinomas, 1 lung squamous cell carcinoma) were inoculated into nude mice to establish in vivo invasion and metastasis models; immunohistochemistry and Western Blot were used. Methods were used to detect the expression of various factors in the uPA system in xenograft tumors. RESULTS: The invasion and metastasis capacity of the giant cell carcinoma cells formed by lung giant cell carcinoma cells was the strongest. Both immunohistochemistry and Western Blot showed that the factors of uPA system were comparable in the cytoplasm and membrane of four tumor cells. The expression of the four factors in the transplanted tumors formed by giant cell lung cancer cells was relatively low, and was relatively high in both adenocarcinoma xenografts. Conclusion : The four factors in the uPA system exist in the form of multiple complexes and monomers in the xenografts; the expression levels of these factors cannot accurately reflect the differences in the invasion and metastasis ability of the xenografts in nude mice.
