目的 :观察具有益气养阴、活血通络作用的中药验方糖肾康对实验性糖尿病大鼠早期肾脏病变的作用及肾脏组织中转化生长因子 β1(TGF - β1)表达的影响。方法 :以胰岛 β细胞毒剂链脲佐菌素 (STZ)诱导产生大鼠糖尿病模型 ,将模型大鼠随机分成糖尿病组、糖肾康治疗组、卡托普利对照组 ,连续灌胃给药 4周 ,观察治疗前后血糖变化 ,尿微量白蛋白排泄 ,血、尿 β2 -MG水平 ,肾脏电镜超微结构及肾脏组织中TGF - β1的表达。结果 :糖肾康可降低糖尿病大鼠肾重 /体重比值 (P <0 .0 5 ) ;糖尿病模型大鼠尿微量白蛋白排泄明显减少 ,血 β2 -MG水平降低 ,尿β2 -MG含量减少 ,减轻 /延缓电镜下肾脏超微结构损害 ,抑制鼠肾组织中TGF - β1表达。结论 :糖肾康对实验性糖尿病肾病有防治作用 ,其机制可能与抑制组织中TGF - β1的表达有关。
Objective : To observe the effect of traditional Chinese medicine prescription Shentangkang, which has the effect of supplementing qi and nourishing yin and promoting blood circulation and collaterals, on early kidney disease in experimental diabetic rats and the expression of transforming growth factor-β1 (TGF-β1) in kidney tissue. METHODS: Diabetic rat model was induced by islet β-cell cytotoxic agent streptozotocin (STZ). Model rats were randomly divided into diabetic group, Tangshen-Kang-treated group, and captopril-treated group. Week, observed blood glucose changes before and after treatment, urinary microalbumin excretion, blood, urine β2-MG levels, ultrastructure of the kidney electron microscope and kidney tissue TGF-β1 expression. Results: Tang Shenkang can reduce the ratio of kidney weight to body weight in diabetic rats (P < 0.05). Diabetic model rats’ urinary albumin excretion was significantly reduced, blood β2-MG levels were decreased, and urinary β2-MG content was decreased. Reduce/delay ultrastructural damage of kidney under electron microscope and inhibit TGF-β1 expression in rat kidney. Conclusion: Tang Shenkang has preventive and therapeutic effects on experimental diabetic nephropathy. The mechanism may be related to inhibiting the expression of TGF-β1 in tissues.
