目的研究3-溴丙酮酸对SWⅢ-C细胞增殖和凋亡的影响。方法利用透射电镜、MTT法、流式细胞术、免疫组化等方法检测3-溴丙酮酸作用于SWⅢ-C细胞后,其细胞形态、超微结构的改变;细胞增殖、细胞周期分布和细胞凋亡率的变化以及Bcl-2、c-myc和mtp53蛋白表达的变化。结果 3-溴丙酮酸可抑制SWⅢ-C细胞的增殖,浓度为25μg·mL-1时抑制作用最明显;3-溴丙酮酸可有效诱导SWⅢ-C细胞凋亡,浓度为25μg·mL-1效果最明显;3-溴丙酮酸可下调SWⅢ-C细胞Bcl-2、c-myc和mtp53蛋白的表达。结论 3-溴丙酮酸可抑制SWⅢ-C细胞增殖并诱导其凋亡,其机制可能与下调Bcl-2、c-myc和mtp53的表达有关。
Objective To investigate the effects of 3-bromopyruvic acid on the proliferation and apoptosis of SW III-C cells. Methods The morphological and ultrastructural changes of SWⅢ-C cells were detected by transmission electron microscopy, MTT assay, flow cytometry and immunohistochemistry. The cell proliferation, cell cycle distribution, The change of apoptosis rate and the change of Bcl-2, c-myc and mtp53 protein expression. Results 3-bromopyruvic acid could inhibit the proliferation of SWⅢ-C cells. The inhibitory effect was most obvious when the concentration was 25μg · mL-1. 3-Brpyruvate could effectively induce SWⅢ-C cell apoptosis at a concentration of 25μg · mL -1 3-bromopyruvic acid can down-regulate the expression of Bcl-2, c-myc and mtp53 in SWⅢ-C cells. Conclusion 3-bromopyruvic acid can inhibit the proliferation and induce the apoptosis of SW III-C cells. The mechanism may be related to the down-regulation of the expression of Bcl-2, c-myc and mtp53.
