目的 :检测针对端粒酶的反义寡核苷酸 (AS -ODN)对鼻咽癌HNE - 1细胞株bcl- 2及c -myc基因表达的影响。方法 :用PCR -ELISA法检测细胞内端粒酶活性。用免疫细胞化学法检测bcl- 2和c -myc基因的表达。 结果 :鼻咽癌细胞株HNE - 1与 2 0 μmol/L反义寡核苷酸共同孵育 1天 ,对其端粒酶活性抑制率为 6 1.6 %。而同浓度正义寡核苷酸 (S -ODN)对细胞端粒酶活性无抑制作用。 2 0 μmol/L反义寡核苷酸作用 2天后鼻咽癌细胞c -myc基因表达HSCORE评分由 3.6 5下降为 1.98,而bcl- 2基因表达HSCORE评分无明显变化。结论 :该反义寡核苷酸可抑制鼻咽癌细胞端粒酶活性 ,此作用有序列特异性。该反义寡核苷酸抑制了鼻咽癌细胞c-myc基因的表达 ,而对bcl- 2基因表达无明显影响。
OBJECTIVE: To detect the effect of antisense oligonucleotide targeting telomerase (AS-ODN) on the gene expression of bcl-2 and c-myc in nasopharyngeal carcinoma HNE - 1 cell line. Methods: Telomerase activity was detected by PCR-ELISA. Immunocytochemistry was used to detect the expression of bcl-2 and c-myc genes. Results: The nasopharyngeal carcinoma cell line HNE - 1 incubated with 20 μmol / L antisense oligonucleotide for 1 day inhibited the telomerase activity of 6. The same concentration of sense oligonucleotide (S-ODN) on cell telomerase activity was not inhibited. The HSCORE score of c-myc gene in nasopharyngeal carcinoma cells decreased from 3.6 5 to 1.98 after 2 μmol / L antisense oligonucleotide treatment for 2 days, but there was no significant change in bcl-2 gene expression HSCORE score. Conclusion: The antisense oligonucleotide can inhibit the telomerase activity in nasopharyngeal carcinoma cells. The effect is sequence-specific. The antisense oligonucleotide inhibited the expression of c-myc gene in nasopharyngeal carcinoma cells, but had no significant effect on bcl-2 gene expression.
