目的 :建立高效液相色谱法测定血浆中加替沙星浓度。方法 :血浆用二氯甲烷提取 ,色谱柱为ZorbaxC18(4.6mm×2 5 0mm) ,流动相为 0 .0 1mol·L-1磷酸二氢钾 甲醇 三乙胺 (6 5∶34.8∶0 .2 ) ,检测波长 2 92nm ,流速 1.0ml·min-1,以司帕沙星为内标。结果 :加替沙星浓度线性范围 0 .0 1~ 5 μg·ml-1,最低检测浓度 0 .0 1μg·ml-1,绝对回收率为 85 .1% ,相对回收率为10 0 .6 %。日内 ,日间RSD分别为 3.0 %和 3.7%。结论 :该方法适用于人体药物动力学研究。
Objective: To establish a method for the determination of gatifloxacin in plasma by high performance liquid chromatography. Methods: The plasma was extracted with dichloromethane. The column was Zorbax C18 (4.6 mm × 250 mm) and the mobile phase was 0.1 mol·L -1 potassium dihydrogen phosphate methanol triethylamine (6 5:34.8:0.2 ), Detection wavelength 2 92nm, flow rate 1.0ml · min-1, with sparfloxacin as internal standard. Results: The linear range of gatifloxacin was 0.0 1 ~ 5 μg · ml-1, the lowest detection limit was 0.0 1 μg · ml-1, the absolute recovery was 85.1% and the relative recovery was 100.6 %. Days, day RSD were 3.0% and 3.7%. Conclusion: This method is suitable for human pharmacokinetic studies.
