目的建立基于组织培养技术的山大颜种子无菌萌发方法。方法观察、采集、记录山大颜果实、种子特征;以不同采收时期新鲜剥去种皮的种子为实验材料,分别进行无菌萌发实验,考察山大颜种子最佳采收时期;以剥去种皮的山大颜成熟种子为外植体,筛选适宜的外植体消毒时间;考察山大颜种子在4种不同培养基上的萌发情况;采用L9(34)正交试验设计法考察6-苄氨基腺嘌呤(6-BA)、赤霉素(GA)、α-萘乙酸(NAA)3种激素对种子萌发的影响。结果 9月份采收的山大颜种子活性较强,可以用作实验材料;2.0 g·L~(-1)升汞溶液浸泡时间2 min的种子生长率高、污染率较小,适合无菌培养;山大颜种子萌发的最优培养基为MS+2.0 mg·L~(-1)6-BA+0.1 mg·L~(-1)GA+3.0 g·L~(-1)活性炭。结论该方法建立了山大颜种子无菌萌发的适宜条件,有助于提高山大颜的繁殖率,为山大颜种质的人工扩大繁殖提供参考。
Objective To establish a germination-free germination method of Da Yan Yan based on tissue culture technology. Methods To observe, collect and record the fruit and seed characteristics of Shannandan, take the seeds of freshly peeled seed coat from different harvesting times as the experimental materials, and conduct germicidal germination experiment respectively, In order to investigate the germination of Selaginella nigra seeds on four different mediums, the mature seeds of Pinus densiflora were used as explants to screen the suitable disinfection time of explants. The L9 (34) orthogonal design was used to investigate the effects of 6-benzylamino Effects of adenosine (6-BA), gibberellin (GA) and α-naphthaleneacetic acid (NAA) on seed germination. Results Seeds collected in September were more active and could be used as experimental materials. Seedlings with 2.0 g · L -1 mercury soaking time of 2 min had higher seed growth rate and lower pollution rate, which was suitable for aseptic culture The optimum medium for the seed germination of P. dahurica was MS + 2.0 mg · L -1 6-BA + 0.1 mg · L -1 GA + 3.0 g · L -1 activated carbon. Conclusion This method established suitable conditions for the germination of A. dahurica seeds, and promoted the reproduction rate of Da Yan. It provided a reference for the artificial propagation of Da Yan Yan.
