目的采用血清药理学方法,研究平肺口服液对人胚肺成纤维细胞增殖及诱导细胞凋亡的作用。方法平肺口服液10g/kg和20g/kg分别灌服大鼠,第1~7天每日一次,第8~10天每日两次,于第11天早上末次灌服后2h腹主动脉无菌采血制备含药血清。采用噻唑蓝(MTT)比色法和流式细胞术分别检测平肺口服液含药血清对人胚肺成纤维细胞(CCC-HPF-1)增殖、细胞凋亡和细胞周期的影响。结果平肺口服液20g/kg灌服所得大鼠含药血清作用96h和120h分别能抑制CCC-HPF-1细胞增殖(P<0.01);流式细胞术检测表明该含药血清作用96h能诱导CCC-HPF-1细胞发生凋亡(P<0.05)和G1/G0期阻滞(P<0.05)。结论平肺口服液能抑制肺成纤维细胞增殖,诱导细胞凋亡以及使细胞周期阻滞于G1/G0期,这可能有助于其预防放射性肺损伤。
Objective To study the effects of Pingfei Oral Liquid on the proliferation and apoptosis of human embryonic lung fibroblasts by serum pharmacology. Methods Pingfei oral liquid 10g / kg and 20g / kg were given to rats on the 1st to the 7th day once a day, on the 8th to 10th day twice daily on the 11th morning 2 hours after the last oral administration of the abdominal aorta Preparation of aseptic blood serum containing medicine. The effects of Pingfei oral liquid-containing serum on the proliferation, apoptosis and cell cycle of human embryonic lung fibroblasts (CCC-HPF-1) were detected by MTT assay and flow cytometry. Results The rat serum containing 20g / kg of Pingfei oral liquid could inhibit the proliferation of CCC-HPF-1 cells at 96h and 120h, respectively (P <0.01). The results of flow cytometry showed that the drug-containing serum could induce 96h Apoptosis (P <0.05) and G1 / G0 arrest (P <0.05) were observed in CCC-HPF-1 cells. Conclusion Pingfei oral liquid can inhibit the proliferation of lung fibroblasts, induce cell apoptosis and arrest the cell cycle in G1 / G0 phase, which may be helpful for the prevention of radiation-induced lung injury.
