目的:研究灯盏生脉胶囊对博莱霉素诱导的小鼠肺纤维化的干预作用。方法:将40只C57BL/6雌性小鼠随机分为盐水对照组(NS)、模型组(Model)、灯盏生脉组(DZSM)和N-乙酰半胱氨酸阳性对照组(N-AC),NS组气管滴注0.1 mL生理盐水,其余3组按5 mg.kg-1剂量滴注博莱霉素溶液复制肺纤维化模型,以后每天分别给予生理盐水、灯盏生脉溶液(222mg.kg-1.d-1)、N-乙酰半胱氨酸溶液(489.6 mg.kg-1)ig治疗,连续给药21 d。实验过程中隔天记录小鼠的生存状态及体重变化,在完成给药后观察肺顺应性和肺阻力的改变状况。留取肺泡灌洗液检测细胞转化生长因子-β(TGF-β)、肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)含量,观察博莱霉素引起的肺炎症改变;留取肺组织,测定肺组织中羟脯氨酸含量,并分别行HE染色和Masson’s染色,观察肺组织的炎症变化和纤维化程度。结果:给予博莱霉素后,各组小鼠体重显著下降,进食减少,肺阻力升高(P<0.05),肺泡灌洗液中TGF-β,TNF-α,IL-1β含量显著升高(P<0.05);灯盏生脉胶囊干预组小鼠体重下降减缓,肺阻力显著下降(P<0.05),肺泡灌洗液中TGF-β,TNF-α,IL-1β含量明显降低(P<0.05);肺组织的病理切片显示肺部炎症及肺纤维化程度均有不同程度的减轻。结论:灯盏生脉胶囊能改善博莱霉素诱导的肺纤维化小鼠的虚弱状态,增强小鼠肺顺应性,降低肺阻力,抑制小鼠的肺组织炎症反应,减轻肺纤维化程度。
Objective: To study the intervention of Dengzhan Shengmai Capsule on bleomycin-induced pulmonary fibrosis in mice. Methods: Forty C57BL / 6 female mice were randomly divided into saline control group (NS), model group, DZSM group and N-acetyl cysteine positive control group (N-AC) , NS group tracheal instillation of 0.1 mL saline, and the remaining 3 groups were treated with 5 mg.kg-1 bleomycin bleomycin solution for pulmonary fibrosis model, after each day were given saline, Dengzhan Shengmai solution (222mg.kg -1.d-1), N-acetyl cysteine solution (489.6 mg.kg-1) ig treatment, continuous administration of 21 d. The survival status and body weight changes of mice were recorded every other day during the experiment, and the changes of lung compliance and lung resistance were observed after completing the administration. The contents of transforming growth factor-β (TGF-β), tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) were detected by the method of alveolar lavage and the inflammatory changes induced by bleomycin The lung tissue was collected and the content of hydroxyproline in lung tissue was determined. HE staining and Masson’s staining were performed respectively to observe the changes of lung inflammation and fibrosis. Results: After bleomycin was administered, the body weight of mice in each group decreased significantly, the food intake decreased and the lung resistance increased (P <0.05). The contents of TGF-β, TNF-α and IL-1β in bronchoalveolar lavage fluid were significantly increased (P <0.05). The body weight of the mice treated with Dengzhan Shengmai Capsule decreased slowly and the lung resistance decreased significantly (P <0.05). The content of TGF-β, TNF-α and IL- 0.05). The pathological sections of lung tissue showed lung inflammation and degree of pulmonary fibrosis were relieved to varying degrees. Conclusion: Dengzhan Shengmai Capsule can improve bleomycin-induced pulmonary fibrosis in mice with weak state, enhance lung compliance, reduce lung resistance, inhibit lung inflammation in mice and reduce the degree of pulmonary fibrosis.
