目的观察大鼠创伤性脑损伤后损伤区周围皮质不同时点聚二磷酸腺苷核糖多聚酶(PARP)阳性细胞的表达变化。方法采用大鼠自由落体脑损伤模型,伤后2、6、12、24、72、168 h取脑切片,行PARP、β-Tubline-Ⅲ、Hoechst荧光检测。结果损伤区周围于伤后2 h即可观察到PARP阳性细胞,主要出现在大脑皮质第3层,6 h数量达到第1个高峰;后PARF阳性细胞出现部位逐渐向深部迁移,至伤后72 h数量达到第2个高峰,表达部位主要位于大脑皮质第5层。伤后各时点均可观察到部分PARP、β-TubulinⅢ双标阳性细胞,其核hoechst着色不均匀,表达变化趋势与PARP阳性细胞基本一致。结论大鼠创伤性脑损伤后损伤区周围PARP阳性细胞数量的变化与伤后时程有关,表达部位逐渐远离损伤区,PARP阳性细胞部分为凋亡的神经元。
Objective To observe the changes of the expression of PARP positive cells in the cortex around the injured area after traumatic brain injury in rats. Methods The rat model of free fall injury was established. Brain sections were taken at 2,6,12,24,72 and 168 h after injury. PARP, β-Tubline-Ⅲ and Hoechst fluorescence were detected. Results PARP positive cells were found in the injured area around 2 h after injury, mainly in the third layer of cerebral cortex, reaching the first peak in 6 h. After PARF positive cells gradually migrated to the deeper part, h number reached the second peak, the expression of the site mainly located in the fifth layer of the cerebral cortex. At each time after injury, some PARP and β-TubulinⅢ double positive cells were observed. The nuclear hoechst staining was not uniform, and the change trend was consistent with that of PARP positive cells. Conclusions The changes of PARP positive cells in traumatic brain injury after traumatic brain injury are related to the time course of injury. The expression of PARP is gradually away from the damaged area and PARP positive cells are apoptotic neurons.
