目的:研究人肾细胞癌(RCC)原代细胞生物学特性及其癌基因的表达情况。方法:应用改良的原代肿瘤细胞分离法分离培养7例人RCC原代细胞。应用染色体核型分析和免疫组化方法,分析原代细胞核型以及癌基因与抑癌基因的表达。结果:用本法培养的原代细胞比其他常规方法所得细胞既多且纯。所培养的人RCC原代细胞的癌基因表达分析发现bcl-2,c-erbB-2,c-met,表皮生长因子受体(EGFR)有轻度至中度表达,而p53没有表达。结论:人RCC原代培养及其癌基因表达检测,为应用原代RCC所开展exvivo的RCC基因治疗打下了初步的基础。
Objective: To study the biological characteristics of primary human renal cell carcinoma (RCC) cells and the expression of their oncogenes. Methods: Seven primary human RCC cells were isolated and cultured using a modified primary tumor cell isolation method. Karyotype analysis and immunohistochemistry were used to analyze the primary cell karyotypes and the expression of oncogenes and tumor suppressor genes. Results: Primary cells cultured by this method were more pure and pure than those obtained by other conventional methods. The analysis of oncogenes of cultured primary human RCC cells showed that mildly to moderately expressed bcl-2, c-erbB-2, c-met, and epidermal growth factor receptor (EGFR) were expressed, while p53 was not expressed. Conclusion: The primary culture of human RCC and the detection of its oncogene expression laid the foundation for the RCC gene therapy of exvivo using primary RCC.
