目的:研究pAdKDR-tk自杀基因系统对鼻咽癌裸鼠中肿瘤血管内皮细胞的作用。方法:建立人鼻咽癌裸鼠模型,将16只成瘤鼠随机分成AdKDR-tk/丙氧鸟苷(GCV)组(治疗组)和AdKDR-tk/PBS组(对照组),每组8只。观察治疗前后肿瘤体积变化、病理组织形态学及用免疫组织化学的方法观察肿瘤微血管密度(MVD)及血管内皮生长因子(VEGF)表达。结果:治疗组在前6d肿瘤缓慢增长,第6天后肿瘤生长受到抑制,移植瘤体积逐渐缩小,而对照组瘤体体积持续迅速增长。治疗后第6、9、14天治疗组瘤体均明显小于对照组(均P<0.01)。治疗组鼻咽癌细胞形态与对照组无明显区别,但瘤组织中有大量的细胞坏死灶;对照组MVD6.63±1.41,治疗组MVD3.50±0.93,组间差异有统计学意义(P<0.05);对照组比治疗组具有较高VEGF阳性表达,2组间VEGF表达的差异具有统计学意义。结论:AdKDR-tk/GCV系统具有抑制裸鼠鼻咽癌肿瘤血管增生及肿瘤生长的作用。
Objective: To investigate the effect of pAdKDR-tk suicide gene system on tumor vascular endothelial cells in nasopharyngeal carcinoma nude mice. Methods: The nude mice model of human nasopharyngeal carcinoma was established. Sixteen tumor-bearing mice were randomly divided into AdKDR-tk / ganciclovir (GCV) group (treatment group) and AdKDR-tk / PBS group only. The changes of tumor volume, histopathology and immunohistochemistry were observed before and after treatment to observe the expression of tumor microvessel density (MVD) and vascular endothelial growth factor (VEGF). Results: In the first 6 days, the tumor in the treatment group grew slowly. After 6 days, the tumor growth was inhibited. The volume of the tumor was gradually reduced, while the volume of the tumor in the control group continued to increase rapidly. On the 6th, 9th and 14th days after treatment, the tumors in the treatment group were significantly smaller than those in the control group (all P <0.01). There was no significant difference between the treatment group and the control group, but there were a large number of necrosis in the tumor tissue. The MVD in the control group was 6.63 ± 1.41, the MVD in the treatment group was 3.50 ± 0.93, and the difference was statistically significant (P <0.05). The control group had higher VEGF expression than the treatment group, and the difference of VEGF expression between the two groups was statistically significant. Conclusion: AdKDR-tk / GCV system can inhibit the proliferation and tumor growth of nasopharyngeal carcinoma in nude mice.
