目的:通过观察补阳还五汤对高血压前期大鼠胸主动脉NF-κB信号通路的影响,探讨其对高血压前期血管炎症损伤的保护机制。方法:选择7周龄盐抵抗大鼠(SS)与盐敏感大鼠(DS),随机分成正常组、模型组、模型给药组和正常给药组,每组各10只。各组大鼠以8%Na Cl高盐饲料喂养,期间采用智能无创血压计监测大鼠尾动脉收缩压,造模成功之后,予以补阳还五汤灌胃治疗,每日1次,连续治疗5周。Elisa法检测血清中TNF-α含量;荧光定量PCR法检测胸主动脉TRAF-6、IKK、IκB、NF-κBp65的m RNA表达。结果:与正常组比较,模型组大鼠TNF-α的含量明显升高,TRAF-6、IKK、NF-κB的m RNA表达显著升高,IκB的m RNA表达明显降低;与模型组比较,模型给药组TNF-α的含量明显降低,TRAF-6、IKK、NF-κB的m RNA表达显著降低,IκB的m RNA表达明显升高。结论:补阳还五汤通过调节TNF-α/NF-κB炎症信号通路,减轻炎症反应,保护血管,从而延缓高血压前期的发展进程。
Objective: To observe the effect of Buyang Huanwu Decoction on NF-κB signal pathway in the thoracic aorta of rats with prehypertension and to explore the protective mechanism of Buyang Huanwu decoction on vascular inflammation injury in the prehypertension. Methods: Salt resistance rats (SS) and salt sensitive rats (DS), 7 weeks old, were selected and randomly divided into normal group, model group, model group and normal group. The rats in each group were fed with high salt diet of 8% NaCl, the systolic pressure of caudal artery was monitored by intelligent noninvasive sphygmomanometer. After the model was established successfully, the rats were treated with Buyang Huanwu Decoction intragastrically once daily for continuous treatment 5 weeks. The content of TNF-α in serum was detected by Elisa method. The m RNA expression of TRAF-6, IKK, IκB and NF-κBp65 in the thoracic aorta was detected by fluorescence quantitative PCR. Results: Compared with the normal group, the content of TNF-α in the model group was significantly increased, the expression of m RNA of TRAF-6, IKK and NF-κB was significantly increased and the expression of m RNA of IκB was significantly decreased. Compared with the model group, The content of TNF-α in the model group was significantly decreased, the m RNA expression of TRAF-6, IKK and NF-κB was significantly decreased, and the m RNA expression of IκB was significantly increased. Conclusion: Buyang Huanwu Decoction can delay the progression of prehypertension by regulating the signaling pathway of TNF-α / NF-κB, relieving inflammation and protecting the blood vessels.
