滋阴平肝法降血压及逆转左室肥厚的实验研究

目的观察滋阴平肝法降血压与逆转左室肥厚的效应。方法自发性高血压大鼠(SHR)24只随机分为中药组、西药组、空白组和模型组,中药组给予滋生青阳片;西药组给予卡托普利片;空白组给予蒸馏水;模型组实验开始即日处死。Wistar大鼠6只,实验开始即日处死,处死前进行有关指标检测,与模型组比较,证明高血压左室肥厚模型成立。中药组、西药组、空白组动物观察时间均为3个月。观察指标:鼠尾动脉收缩压(SBP);左室重量(LVM)及左室重量/体重比(LVM/BW);心肌羟脯氨酸(HCA)、心肌胶原浓度(JYC)、心肌胶原含量(JY);血清Ⅲ型前胶原氨基端肽(PⅢNP)、血清透明质酸(HA)等。结果SHR大鼠SBP显著升高;LVM、LVM/BW、HCA、JYC、JY、PⅢNP、HA明显增加,表明SHR大鼠存在高血压左室肥厚。中药组、西药组与空白组比较,SBP显著降低,中药组与西药组比较,其降压作用缓和、平稳。中药组、西药组分别与空白组比较,LVM、LVM/BW、HCA、JYC、JY均明显低于空白组。中药组与西药组比较,中药组HCA、JYC明显低于西药组。三组比较,中药组和西药组血清PⅢNP和HA明显低于空白组;中药组PⅢNP、HA明显低于西药组。结论滋生青阳片与卡托普利片对SHR大鼠有良好的降压和逆转左室肥厚的作用,能明显改善SHR大鼠心肌纤维化。滋阴平肝法及其组方滋生青阳片具有缓和、平稳的降压特点,在减缓心肌纤维化进程、促进左室肥厚逆转方面显示出于优于西药的趋势。 Objective To observe the effect of Ziyin Pinggan method in reducing blood pressure and reversing left ventricular hypertrophy. Methods Twenty-four spontaneously hypertensive rats (SHR) were randomly divided into traditional Chinese medicine group, western medicine group, blank group and model group. Chinese medicine group was given Qingsheng Qingyang Tablet; western medicine group was given captopril; blank group was given distilled water; The group experiment was started on the same day. Six Wistar rats were sacrificed on the same day of the experiment and were tested before sacrifice. Compared with the model group, the model of left ventricular hypertrophy was established. The observation time for the Chinese medicine group, western medicine group and blank group were all 3 months. Observation indexes: SBP of rat tail artery; Left ventricular weight (LVM) and left ventricular mass/weight ratio (LVM/BW); Myocardial hydroxyproline (HCA), Myocardial collagen concentration (JYC), Myocardial collagen content (JY); serum type III procollagen amino-terminal peptide (PIIINP), serum hyaluronic acid (HA) and the like. Results SBP significantly increased in SHR rats; LVM, LVM/BW, HCA, JYC, JY, PIIINP, HA increased significantly, indicating that SHR rats have left ventricular hypertrophy. Compared with the blank group, Chinese medicine group, Western medicine group, SBP significantly reduced, Chinese medicine group and Western medicine group, its antihypertensive effect is moderate, stable. Compared with the blank group, the LVM, LVM/BW, HCA, JYC, and JY were significantly lower in the Chinese medicine group and the western medicine group than in the blank group. Compared with the Western medicine group, Chinese medicine group HCA, JYC significantly lower than the Western medicine group. Compared with the three groups, serum PIIINP and HA were significantly lower in the Chinese medicine group and the western medicine group than in the blank group; PIIINP and HA in the Chinese medicine group were significantly lower than those in the western medicine group. Conclusion The results showed that the combination of Qingyang Tablets and Captopril Tablets can effectively reduce blood pressure and reverse left ventricular hypertrophy in SHR rats, and can significantly improve myocardial fibrosis in SHR rats. Ziyin Pinggan Method and its combination of Fangshen Qingyang Tablets have a mild and steady blood pressure decompression feature, showing a tendency to be superior to western medicine in slowing the progression of myocardial fibrosis and promoting the reversal of left ventricular hypertrophy.
