目的观察微小RNA-424-3p和5p在LNCaP中的表达,并比较其对LNCaP生物学行为的影响。方法应用实时荧光定量聚合酶链式反应(FQ-PCR)检测miR-424-3p和5p的表达量。利用细胞计数盒,平板克隆试验,Transwell小室,流式细胞仪检测LNCaP的增殖、迁移及细胞周期变化。结果 miR-424-3p和5p在LNCaP中的表达量高于RWPE-1;miR.-424-3p和5p下调后,LNCaP的增殖、迁移受到抑制,早期凋亡细胞比例增加,S期细胞比例增加。结论miR-424-3p和5p在LNCaP中高表达且5p升高更为显著,但比较两者对LNCaP生物学行为影响的大小,差异无统计学意义(P>0.05),提示miR-3p和5p对肿瘤细胞的生物学行为影响大小与其丰度并不一定成正比例关系。
Objective To observe the expression of microRNA-424-3p and 5p in LNCaP and to compare their effects on the biological behavior of LNCaP. Methods Real-time fluorescent quantitative polymerase chain reaction (FQ-PCR) was used to detect the expression of miR-424-3p and 5p. The proliferation, migration and cell cycle changes of LNCaP were detected by cell counting kit, plate clone assay, Transwell chamber and flow cytometry. Results The expression of miR-424-3p and 5p in LNCaP was higher than that in RWPE-1. After miR-424-3p and 5p were down-regulated, the proliferation and migration of LNCaP were inhibited, the proportion of early apoptotic cells increased and the proportion of S phase cells increase. Conclusion The expression of miR-424-3p and 5p in LNCaP is higher than that in LNCaP, but the difference between them is not statistically significant (P> 0.05), suggesting that miR-3p and 5p The biological behavior of tumor cells, the size and abundance is not necessarily proportional relationship.
