目的 检测昆明山海棠碱 (THH碱 )诱导JurkatT淋巴瘤细胞的凋亡效应。方法 THH碱多剂量多时间点诱导JurkatT淋巴瘤细胞细胞后 ,采用Annexin V Fluorescence标记膜外翻磷脂酰丝氨酸 (Phosphatidylserine ,PS)的凋亡细胞 ,同时荧光素PI标记坏死细胞 ,流式细胞术检测分别检测其阳性细胞率。同时采用判断凋亡发生的“金标准”———形态学观察方法观察其超微结构的改变。结果 THH碱诱导后磷脂酰丝氨酸外翻的细胞显著增加 ,有明显的时间和剂量相关性 ,超微结构观察可见大量细胞明显发生细胞体积缩小、核固缩、胞浆浓缩、凋亡小体形成等凋亡典型的形态特征。结论 THH碱能非常显著地诱导JurkatT淋巴瘤细胞发生凋亡
Objective To detect the apoptosis of Jurkat T lymphoma cells induced by THH in Kunming.
