采用循环伏安法对半乳糖氧化酶(GAO)进行了生物电化学特性的研究。实验结果表明,由于半乳糖氧化酶的酶活性中心深埋在酶内部,所以该酶在铂电极上的直接电化学行为较难观察到,而二茂铁甲醇作为介体能明显地改善酶活性中心的电子传递,加快电子传递速率。且测定了半乳糖氧化酶与二茂铁甲醇之间的反应速度常数Ks为1.2×10~5L·mol~(-1)·s~(-1)。
Cyclic voltammetry was used to study the bioelectrochemical properties of galactose oxidase (GAO). The experimental results show that the direct electrochemical activity of the enzyme on platinum electrode is hard to be observed because the enzyme activity center of galactose oxidase is deeply buried in the enzyme, and ferrocene methanol as the mediator can obviously improve the enzyme activity center The electron transfer speed up the electron transfer rate. The reaction rate constant Ks between galactose oxidase and ferrocene methanol was 1.2 × 10 ~ 5L · mol -1 s -1.
