目的:观察促排卵所致低容受性小鼠子宫内膜的变化及其中医辨证特性,探讨当归散对子宫内膜中IL-1β表达的影响。方法:采用曲普瑞林+HMG+HCG长周期促排卵方法制备小鼠低容受性子宫模型,造模同时分别预防性给予当归散、当归-黄芩以及当归散去当归-黄芩供试液,分别于排卵后24h和48h分离子宫,观察各组子宫内膜形态变化,并测定内膜中IL-1β表达水平。结果:(1)子宫内膜形态:模型组子宫内膜增厚,间质发育异常,血管发育不良,与正常组差异显著。当归散组及归-芩组内膜偏厚,但血管发育较好,与正常组相近;去归-芩组与模型组接近。(2)IL-1β表达:排卵后24h,模型组IL-1β表达与正常组无明显差异;归-芩组IL-1β值低于正常组水平(P<0.05)。48h时,当归散组IL-1β值无明显变化,其余各组表达均较24h时显著升高(P<0.05或0.01);其中模型组和去归-芩组IL-1β表达上调明显,高于正常组(P<0.05);当归散组IL-1β值显著低于模型组(P<0.05),与正常组无差异。结论:促排卵药物引起的动物子宫内膜发育不良,其显微变化与中医血虚、血瘀及热证病变吻合;当归散能改善造模引起的子宫内膜发育不良;当归散能调节子宫内膜的IL-1β的适度表达;方中当归-黄芩配伍体现了当归散和血清热的核心立法思想。
OBJECTIVE: To observe the changes of endometrium in low tolerance mice induced by ovulation induction and their TCM syndrome differentiation characteristics, and to explore the effect of Danggui Powder on the expression of IL-1β in endometrium. Methods: The low tolerance uterus model was established by triptorelin + HMG + HCG long-term ovulation induction method. At the same time, the model of Angelicae Sinensis - Radix Scutellariae and Radix Angelicae Sinensis - The uterus was isolated at 24h and 48h after ovulation, the morphological changes of endometrium in each group were observed, and the expression of IL-1β in the intima was measured. Results: (1) Endometrial morphology: Thickening of endometrium, interstitial dysplasia and vascular dysplasia in model group showed significant difference from normal group. Angelica powder group and Gui - Qin group endometrial partial thick, but good vascular development, and normal group is similar to the - Qin - Qin group and the model group close. (2) IL-1βexpression: At 24 hours after ovulation, the expression of IL-1β in the model group had no significant difference compared with the normal group; the IL-1β level in the Gui-Qin group was lower than that of the normal group (P <0.05). At 48h, there was no significant change in the IL-1β level in Angelica Siniuensis group, while the expression in other groups was significantly higher than that in 24h (P <0.05 or 0.01) IL-1β in Danggui Powder group was significantly lower than that in model group (P <0.05), but not in normal group. Conclusion: Ovulation drugs cause animal endometrial dysplasia, and its micro-changes and TCM blood deficiency, blood stasis and heat syndrome anastomosis; Angelica scattered to improve the modeling-induced endometrial dysplasia; Angelica powder to regulate the uterus Moderate expression of IL-1β in the intima; compatibility of Radix Angelicae Sinensis and Radix Scutellariae reflects the core legislative thoughts of Angelica sinensis and serum heat.
