目的:筛选贞清胶囊的水提醇沉最佳工艺条件。方法:高效液相色谱法测定水煎液和醇沉液中马钱苷的含量;以马钱苷提取率和干膏率为评价指标,采用正交设计法考察加水量、水提时间、水提次数等因素对水提工艺的影响;考察醇沉前药液相对密度、醇沉浓度、醇沉时间三因素对醇沉工艺的影响。结果:最佳水提工艺条件为加入8倍量水,提取2次,每次1.5h(100℃);最佳醇沉工艺条件即加入乙醇使含醇量达60%,水提液浓缩体积比为1∶1.2,醇沉时间24h。结论:经正交设计筛选贞清胶囊的水提醇沉工艺是科学可行的。
Objective: To screen the optimum conditions of water extraction and alcohol precipitation of Zhenqing capsule. Methods: The content of loganin in decoction and alcohol sediment was determined by high performance liquid chromatography (HPLC). Using the extraction rate of loganin and the rate of dry cream as evaluation indexes, the orthogonal design method was used to investigate the effects of water addition, water extraction time, The number of times and other factors on the impact of water extraction process; study alcohol precipitation before the relative density, alcohol concentration, alcohol precipitation time three factors on alcohol precipitation process. Results: The optimal conditions for water extraction were adding 8 times the amount of water and extracting for 2 times, each time being 1.5h (100 ℃). The optimum alcohol precipitation conditions were as follows: ethanol was added to make the alcohol content reach 60% Ratio of 1: 1.2, alcohol precipitation time 24h. Conclusion: It is scientifically feasible to screen the water extract and alcohol precipitation of Zhenqing capsule by orthogonal design.
