目的 研究常见病原菌 2 3SrRNA基因序列变异规律 ,为细菌鉴别诊断和相关研究提供科学依据。方法 运用Genbank信息和生物学分析软件 ,并通过PCR扩增 2 3SrRNA基因片段 ,分析不同种属细菌 2 3SrRNA基因的保守序列、变异规律及菌株间种系进化关系。结果 通过基因扩增和生物信息学比较分析 ,获得 2 1个 2 3SrRNA的通用保守序列 ,2 3SrRNA基因保守序列与变异区在种系间呈间隔分布 ,大量变异区呈“马赛克”式分布。种系间进化关系与 1 6SrRNA分析结果一致。结论 2 3SrRNA基因序列变异规律为进行细菌的鉴别诊断提供了重要的理论基础
Objective To study the sequence variation of 23SrRNA gene in common pathogens and provide a scientific basis for the differential diagnosis and related studies of bacteria. Methods Genomic information and biological analysis software were used to amplify the 23SrRNA gene fragment by PCR. The conserved sequence, variation pattern and germline evolutionary relationships among 23SrRNA genes of different species were analyzed. Results A total of 21 conserved sequences of 23SrRNAs were obtained by gene amplification and bioinformatics analysis. The conserved sequences of 23SrRNAs and the variation regions were interspersed among the germlines, and a large number of variation regions were “mosaic”. The phylogenetic relationships were consistent with the results of 1 6S rRNA analysis. Conclusion The variation of 3SrRNA gene sequence provides an important theoretical basis for the differential diagnosis of bacteria
