目的 确定人体内能产生杀菌 /通透性增加蛋白(bactericidal/ permeability- increasing protein,BPI)的细胞群体 ,为研究 BPI在细胞内的定位奠定基础 .方法 分离人外周血获得嗜中性多形核粒细胞 (polymorphonuclear neutrophils,PMN )和单个核细胞 ,PMN,HL - 6 0及单个核细胞分别进行RNA提取 ,逆转录 PCR反应扩增 BPI基因片段和免疫荧光法检测 .结果 逆转录 PCR在 PMN和 HL- 6 0细胞中扩增出BPI基因片段 ,活细胞免疫荧光染色观察到 PMN和 HL- 6 0能够与抗 BPI的单抗所结合而呈阳性 .结论 BPI是中性粒系细胞的特异性产物
