目的:初步评价重组人β防御素3(rHBD3)对附着于义齿软衬硅橡胶材料表面白色念珠菌的抑菌效果。方法:在软衬硅橡胶试件上面接种0.1 mL的白色念珠菌,然后将试件分别浸泡在25μg/mL rHBD3和5.25%NaClO溶液中5、10、30、60 min。将接种过菌液的一面的试件放置在BHI琼脂培养板上15 s。隔天计算菌落数,采用SPSS10.0软件包对数据进行双因素方差分析。利用激光共聚焦显微镜观察rHBD3对白色念珠菌形成的细菌生物膜的作用。结果:白色念珠菌菌落数在rHBD3作用不同时间后,变化有显著差异(P<0.05)。30 min后,rHBD3与5.25%NaClO组间无显著差异。白色念珠菌生物膜在rHBD3作用30 min后,SYTO9/PI染色显示死菌的数量明显增多。结论:rHBD3不仅能够消除软衬表面白色念珠菌等微生物的感染,而且对其形成的细菌生物膜也有很好的抑菌效果。rHBD3用于软衬材料的清洁维护,为预防义齿性口炎的发生提供了实验依据。
OBJECTIVE: To evaluate the antibacterial effect of recombinant human β-defensin 3 (rHBD3) on Candida albicans attached to the surface of denture soft silicone rubber. Methods: 0.1 mL Candida albicans was inoculated on the soft lining silicon rubber specimens, and the specimens were immersed in 25μg / mL rHBD3 and 5.25% NaClO solution for 5, 10, 30, 60 min respectively. The specimen inoculated with the bacterial liquid was placed on a BHI agar plate for 15 seconds. The number of colonies was calculated every other day, and the data were analyzed by two-way ANOVA using SPSS10.0 software package. The effect of rHBD3 on bacterial biofilm formation by Candida albicans was observed by laser confocal microscopy. Results: The number of Candida albicans colonies changed significantly at different time after rHBD3 treatment (P <0.05). After 30 min, there was no significant difference between rHBD3 and 5.25% NaClO groups. Candida albicans biofilms in rHBD3 role 30 min, SYTO9 / PI staining showed a significant increase in the number of dead bacteria. Conclusion: rHBD3 can not only eliminate the infection of microorganisms such as Candida albicans on the soft lining, but also have good antibacterial effect on the bacterial biofilm formed by it. rHBD3 is used for the cleaning and maintenance of soft liner materials, which provides experimental evidence for preventing the occurrence of denture stomatitis.
