目的:探讨双黄连、鱼腥草在细胞水平对鼠肝炎病毒3型(murine hepatitis virus,MHV-3)是否具有抑制作用。方法:细胞培养、蚀斑试验测定病毒滴度;MTT法进行药物毒性试验;蚀斑抑制试验检测双黄连、鱼腥草的抗MHV-3活性。结果:双黄连半数中毒质量浓度(TC50)为6 100μg.m l-1,抗MHV-3病毒的半数抑制质量浓度(IC50)为304.9μg.m l-1;鱼腥草的分别为500、22.06μg.m l-1。治疗指数(TI)双黄连为20.07,鱼腥草为22.67。两种中药成分对MHV-3病毒的抑制作用均存在明显的量效反应关系(P<0.01)。结论:双黄连、鱼腥草在细胞水平有显著的抗MHV-3作用。
Objective: To investigate whether Shuanghuanglian and Houttuynia had inhibitory effects on murine hepatitis virus (MHV-3) at the cellular level. METHODS: Cell culture and plaque assay were used to determine virus titer. MTT assay was used for drug toxicity test. Plaque inhibition assay was used to detect anti-MHV-3 activity of Shuanghuanglian and Houttuynia cordata. Results: The concentration of TC50 of SHL was 6 100 μg.m 1 -1, the IC50 of anti-MHV-3 virus was 304.9 μg.m l -1, and that of Houttuynia was 500, respectively. 22.06 μg.m l-1. The treatment index (TI) was 20.07 for Shuanghuanglian and 22.67 for Houttuynia. There was a significant dose-response relationship between the inhibitory effects of two traditional Chinese medicine components on MHV-3 virus (P<0.01). Conclusion: Shuanghuanglian and Houttuynia had a significant anti-MHV-3 effect at the cellular level.
