目的:建立新疆一枝蒿中的主要有效成分木犀草素与一枝蒿酮酸的高效液相含量测定方法。方法:采用高效液相色谱法测定,Shim packODSC18(4.6 mm×250 mm,5μm);甲醇-0.4%磷酸(45∶55),检测波长350 nm,该条件下测定木犀草素;检测波长241 nm,该条件下测定一枝蒿酮酸。结果:木犀草素在12.80~51.20μg.mL-1范围内,峰面积与进样浓度具有良好线性关系,r=0.999 8,平均回收率为99.92%(n=9);一枝蒿酮酸在9.72~77.76μg.mL-1范围内线形关系也良好,r=0.999 3,平均回收率为99.82%(n=9)。结论:本法专属性强,操作简便,结果准确。适用于检测新疆一枝蒿中木犀草素和一枝蒿酮酸的含量。
OBJECTIVE: To establish a method for the determination of luteolin and glucosyl acid in the main effective components of Artemisia halodendron. METHODS: HPLC was used to determine Shim packODSC18 (4.6 mm×250 mm, 5 μm); methanol-0.4% phosphoric acid (45:55) at a detection wavelength of 350 nm. Luteolin was determined at this condition. The detection wavelength was 241 nm. In this condition, a candidone acid was determined. Results: Luteolin was in the range of 12.80-51.20 μg.mL-1. The peak area was in good linear relationship with the injection concentration. The average recovery was 99.92% (n=9); The linear relationship was also good in the range of 9.72-77.76μg.mL-1, r=0.999 3, and the average recovery was 99.82% (n=9). Conclusion: This law is highly specific, simple and accurate. It is suitable for detecting the content of luteolin and glucosyl acid in Artemisia halodendron.
