本研究检测cmtm5(CKLF-like MARVEL transmembrane domain containing member5)基因在多发性骨髓瘤(MM)骨髓细胞中的表达水平,并探索其与临床特征的关系。应用实时定量逆转录-聚合酶链反应(real-time quantitative RT-PCR,RQ-RT-PCR)技术检测MM患者骨髓细胞、MM细胞系及正常人骨髓细胞cmtm5基因的表达。以cmtm5拷贝数与abl拷贝数的比值表示cmtm5基因的相对表达水平。结果表明,51例初治及难治复发MM患者骨髓细胞cmtm5表达量(0.047±0.062)显著低于20例正常人骨髓细胞cmtm5表达量(0.255±0.333,p<0.01)。ISSⅢ期患者组cmtm5基因表达量(0.034±0.034)显著低于ISSⅠ期患者组的表达量(0.103±0.109,p<0.01)。两种人多发性骨髓瘤细胞系RPMI8226及CZ1的cmtm5基因表达量(为0.014±0.009、0.004±0.006)均低于正常人骨髓细胞。15例治疗有效患者的cmtm5基因表达量与治疗前及复发时相比显著升高(0.020±0.005;0.227±0.038,p<0.01)。cmtm5基因的表达量与骨髓浆细胞比例呈负相关(r=-0.307,p<0.05),而与性别、年龄、血红蛋白水平、M蛋白量、免疫球蛋白分型、IgH是否阳性等没有明显的相关性。结论:多发性骨髓瘤患者骨髓细胞cmtm5基因表达水平降低,且与ISS分期有关,与骨髓浆细胞数呈负相关,此结果说明cmtm5基因在MM的发病机制中发挥一定作用。
This study was aimed to investigate the expression level of cmtm5 (MKLF-like MARVEL transmembrane domain containing member5) gene in multiple myeloma (MM) bone marrow cells and to explore its relationship with clinical features. The real-time quantitative RT-PCR (RQ-RT-PCR) was used to detect the expression of cmtm5 gene in bone marrow cells, MM cells and normal human bone marrow cells in MM patients. The ratio of cmtm5 copy number to abl copy number indicates the relative expression level of cmtm5 gene. The results showed that the expression of cmtm5 in bone marrow cells (0.047 ± 0.062) in 51 newly diagnosed and refractory relapsed MM patients was significantly lower than that in 20 healthy people (0.255 ± 0.333, p <0.01). The expression of cmtm5 gene in ISS stage Ⅲ patients (0.034 ± 0.034) was significantly lower than that in ISS stage Ⅰ patients (0.103 ± 0.109, p <0.01). The expression of cmtm5 in two human multiple myeloma cell lines RPMI8226 and CZ1 (0.014 ± 0.009, 0.004 ± 0.006) were lower than those in normal human bone marrow cells. The expression of cmtm5 in 15 patients with active treatment was significantly higher than that before treatment and at relapse (0.020 ± 0.005; 0.227 ± 0.038, p <0.01). The expression of cmtm5 gene was negatively correlated with the proportion of bone marrow plasma cells (r = -0.307, p <0.05), but not with sex, age, hemoglobin level, M protein level, immunoglobulin type and IgH Correlation. Conclusion: The expression of cmtm5 gene in bone marrow cells of multiple myeloma patients is lower than that of ISS stage and negatively correlated with the number of bone marrow plasma cells. The results suggest that cmtm5 gene plays a role in the pathogenesis of MM.
