目的 探讨体外培养条件下 ,阿司匹林和吲哚美辛对幽门螺杆菌 (Hp)生长的影响 ,以及阿司匹林存在时 ,Hp对几种常用抗生素敏感性的影响。 方法 Hp接种于含不同浓度阿司匹林或吲哚美辛的布氏肉汤中培养 ,测定不同培养时间菌液的吸光度 ,稀释法计数活菌数 ,革兰染色观察细菌形态。琼脂稀释法测定阿司匹林和吲哚美辛的最低抑菌浓度 (MIC)。E 检测法检测Hp对甲硝唑、克拉霉素及羟氨苄青霉素的MIC。结果 阿司匹林和吲哚美辛可抑制Hp的生长 ,这种抑制作用呈剂量依赖性 ,与培养基 pH改变无关。 4 0 0 μg/ml阿司匹林或 10 0 μg/ml吲哚美辛体外培养 4 8h可使Hp完全溶解破坏。阿司匹林和吲哚美辛对Hp的体外MIC90 分别为 5 12和 12 8μg/ml。阿司匹林存在时 ,分别使10 0 % ,75 %及 75 %的Hp对羟氨苄青霉素、甲硝唑及克拉霉素的MIC降低 ,提示阿司匹林可使Hp对上述抗生素的敏感性增高。结论 阿司匹林和吲哚美辛可抑制Hp的生长 ,阿司匹林可提高Hp对甲硝唑、克拉霉素及羟氨苄青霉素的敏感性
Objective To investigate the effects of aspirin and indomethacin on the growth of Helicobacter pylori (Hp) in vitro and the effect of Hp on the susceptibility of several commonly used antibiotics in the presence of aspirin. Methods Hp was inoculated into the broth broth containing different concentrations of aspirin or indomethacin. The absorbance of the broth was measured at different culture times. The number of viable cells was counted by dilution method and the bacterial morphology was observed by Gram stain. Agar dilution method for the determination of aspirin and indomethacin minimum inhibitory concentration (MIC). E detection method for the detection of Hp metronidazole, clarithromycin and amoxicillin MIC. Results Aspirin and indomethacin inhibited the growth of Hp in a dose-dependent manner, which was not related to the pH change of the medium. 4 0 0 μg / ml aspirin or indometacin 100 μg / ml in vitro 48 h can completely dissolve Hp damage. In vitro MIC90 for aspirin and indomethacin for Hp was 5 12 and 12 8 μg / ml, respectively. In the presence of aspirin, the MIC of Hp to amoxicillin, metronidazole and clarithromycin was reduced at 10%, 75% and 75%, respectively, suggesting that aspirin may increase the sensitivity of Hp to these antibiotics. Conclusion Aspirin and indomethacin can inhibit the growth of Hp. Aspirin can increase the sensitivity of Hp to metronidazole, clarithromycin and amoxicillin
