采用 mRNA差异展示技术分离正常和经(60)COγ射线照射的人肠上皮细胞株 HIEC细胞之间的差异表达基因,为进一步揭开电离辐射对胃肠细胞损伤的分子机理奠定基础。从正常的和经γ射线照射的人肠上皮细胞中分离出差异条带共101条,31个为新表达的差异片段,29个为高表达的差异片段, 41个为要表达的差异片段;其中 10个片段属于 D-T(11)G组, 59个片段属于 D-T(11)A组; 32个片段属于 D-T(11)C组。从新表达的差异片段中随机挑取 5个片段作探针,与正常的及照射后的人肠上皮细胞总RNA进行打点杂交,进一步证实呈差异表达。结果表明,101个差异表达基因与γ辐射损伤有关,其中31个新表达基因可能与电离辐射引起正常组织损伤关系更密切。
The mRNA differential display technique was used to separate the differentially expressed genes between normal and irradiated (60) COγ-ray human intestinal epithelial cell line HIEC cells, which laid the foundation for further revealing the molecular mechanism of ionizing radiation on gastrointestinal cell injury. A total of 101 differential bands were isolated from normal and γ-ray irradiated human intestinal epithelial cells, 31 were differentially expressed, 29 highly expressed and 41 differentially expressed. Among them, 10 fragments belong to D-T (11) G group, 59 fragments belong to D-T (11) A group and 32 fragments belong to D-T (11) C group. Five fragments were randomly selected from the newly expressed differential fragments as probes to dot blot hybridization with normal and irradiated human intestinal epithelial cells to further confirm the differential expression. The results showed that 101 differentially expressed genes were associated with γ radiation injury, of which 31 newly expressed genes might be related to normal tissue damage induced by ionizing radiation.
