目的 探讨转化生长因子 -β1 ( TGF-β1 )在人乳头状甲状腺癌和良性腺瘤、桥本氏甲状腺炎及正常组织中的表达差别。方法 利用逆转录多聚酶链反应 ( RT-PCR)及表达峰度定量法 ,检测 4种新鲜甲状腺手术标本组织中 TGF-β1 m RNA的表达。结果 TGF-β1 m RNA在 4种甲状腺组织中的表达均有较高的阳性率 ( >83 .3 % ) ,乳头状甲状腺癌中 TGF -β1 m RNA的表达量较良性腺瘤、桥本氏炎及正常组织均增多 ( P <0 .0 5 ) ,后三者之间则无差别。结论 TGF -β1 m RNA在乳头状甲状腺癌中表达代偿性增强与癌的发生有关 ,且可作为乳头状甲状腺癌的诊断指标之一 ,并辅助鉴别良恶性 ;RT-PCR则为临床上较为快捷、有效的辅助诊断方法
Objective To investigate the expression of transforming growth factor-β1 (TGF-β1) in human papillary thyroid carcinoma and benign adenoma, Hashimoto’s thyroiditis and normal tissues. Methods Reverse transcription-polymerase chain reaction (RT-PCR) and kurtosis assay were used to detect the expression of TGF-β1 mRNA in four fresh thyroid surgical specimens. Results The positive rate of TGF-β1 m RNA expression in all four thyroid tissues was higher than that in normal thyroid tissues (> 83.3%). The expression of TGF-β1 m RNA in papillary thyroid carcinoma was significantly higher than that in benign adenoma, Hashimoto Inflammation and normal tissue were increased (P <0. 05), the latter three no difference between. Conclusion The expression of TGF-β1 m RNA in papillary thyroid carcinoma is compensatory and is related to the occurrence of cancer, and it can be used as a diagnostic index of papillary thyroid carcinoma and help to distinguish benign and malignant; RT-PCR is clinically more Quick and effective diagnostic methods
