目的:研究人脂多糖应答基因(Lrg)这一新分子在正常胎儿组织中的表达分布情况。方法:利用课题组制备的兔抗人Lrg免疫血清,对胎儿心脏、肝脏、肺脏、肾脏、胃、小肠、睾丸和皮肤等进行免疫组化染色;特异性标记lrg目的基因,用D igoxin标记检测试剂盒,对胎儿小肠及睾丸组织进行原位杂交检测。结果:免疫组化实验表明Lrg在几种检测组织内均有表达,表达水平由强到弱依次为胃、睾丸、食管、小肠、肝、肾、心、肺等;原位杂交方法检测表明人lrg-mRNA在睾丸及小肠组织内高表达。结论:人Lrg在正常胎儿组织中有表达,但分布情况有差异,可能是一种新型功能蛋白。
Objective: To study the expression and distribution of Lrg, a novel molecule in normal fetal tissue. Methods: Immunohistochemical staining of fetal heart, liver, lung, kidney, stomach, small intestine, testis and skin was performed by using rabbit anti-human Lrg immune serum prepared by the research group. The gene of lrg was specifically labeled and detected by D igoxin labeling Kit, fetal small intestine and testicular tissue in situ hybridization detection. Results: The results of immunohistochemistry showed that Lrg was expressed in several kinds of tissues. The expression levels of Lrg in stomach, testis, esophagus, small intestine, liver, kidney, heart and lung were in descending order. The in situ hybridization method showed that human lrg-mRNA is highly expressed in testis and small intestine. Conclusion: Human Lrg is expressed in normal fetal tissue but its distribution is different, which may be a new type of functional protein.
