目的研究重组人TNF-α(recomb inant hum an tumor necrosis factor-,αrhTNF-α)对扁桃体B原淋巴细胞(tonsil B lymphob lastoid cellline,TB-LCL)的趋化作用,建立一个模型以研究细胞因子与B细胞相互间的作用。方法用EB病毒(Epste in-Bar virus,EBV)转化扁桃体B细胞建立TB-LCL系,以小室法研究rhTNF-α)对TB-LCL的趋化作用。结果rhTNF-α对TB-LCL有明显趋化作用,并呈剂量依赖关系。这种作用能为抗TNF-α的单抗E6所阻断。结论以上为研究细胞与细胞因子间的相互作用提供了有用而简便的模型。
Objective To investigate the chemotactic effect of recombinant human TNF-α (αrhTNF-α) on tonsil B lymphoblastoid lymphocyte (TB-LCL) and to establish a model to study cytokines Interaction with B cells. Methods Transforming tonsil B cells with Epstein-Barr virus (EBV) to establish a TB-LCL line and to study the chemotactic effect of rhTNF-α on TB-LCL by the chamber method. Results rhTNF-α had obvious chemotaxis to TB-LCL in a dose-dependent manner. This effect can be blocked by anti-TNF-α mAb E6. Conclusions The above provides a useful and easy model for studying the interaction between cells and cytokines.
