目的:采用HPLC法建立紫穗槐根的指纹图谱,并建立同时测定紫穗槐根中毛蕊异黄酮葡萄糖苷、芒柄花苷、毛蕊异黄酮、芒柄花素含量的方法。方法:采用Diamonsil C18色谱柱(250 mm×4.6 mm,5μm);以乙腈(A)-0.2%磷酸溶液(B)为流动相,梯度洗脱;检测波长为260 nm。对采集的10批紫穗槐根进行测定,运用“中药色谱指纹图谱相似度评价软件”(2004A)版进行相似度评价,运用SPSS软件进行聚类分析。结果:建立了紫穗槐根的HPLC指纹图谱及4种成分的含量测定方法。10批紫穗槐根的HPLC指纹图谱共标定16个共有峰,相似度均在0.90以上。结论:该方法建立的紫穗槐根HPLC指纹图谱特征性强、重复性好,为紫穗槐根的鉴定及质量控制提供了参考。
OBJECTIVE: To establish a fingerprint of Amorpha fruticosa L. by HPLC and to establish a method for the simultaneous determination of calluses, monascus glycosides, calycifloron and formononetin in Amorpha fruticosa roots. METHODS: Diamonsil C18 column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of acetonitrile (A) - 0.2% phosphoric acid solution (B) with gradient elution. The detection wavelength was 260 nm. The collected 10 batches of Amorpha fruticosa root were determined, and the similarity was evaluated by using the similarity evaluation software of chromatographic fingerprints of traditional Chinese medicine (2004A), and the cluster analysis was carried out using SPSS software. Results: HPLC fingerprinting of Amorpha fruticosa L. and determination of four components were established. The HPLC fingerprints of 10 batches of Amorpha fruticosa were co-calibrated to 16 common peaks with similarities of more than 0.90. Conclusion: The HPLC fingerprints of Amorpha fruticosa L. established by this method are characterized by strong reproducibility and good repeatability, which provide references for the identification and quality control of Amorpha fruticosa.
