目的:探讨补肾活方联合微囊化细胞移植技术对大鼠生殖内分泌的影响。方法:将50只SD雌性大鼠随机分成5组:空白组(A组)、去卵巢组(B组)、微囊化GC细胞移植组(C组)、低剂量补肾中药+微囊化GC细胞移植组(D组)、高剂量补肾中药+微囊化GC细胞移植组(E组)。采用酶联免疫吸附法(ELISA)检测治疗前后血清雌二醇(E_2)、卵泡刺激素(FSH)及黄体生成素(LH)浓度。结果:微囊化GC细胞移植后,C组大鼠血清E_2水平与B组比较显著升高(P<0.05);FSH、LH水平均明显降低(P<0.01);补肾活血方联合微囊化GC细胞移植后,D、E组E_2水平与C组比较显著升高(P<0.05),FSH、LH水平均明显降低(P<0.05)。结论:与单纯移植微囊化GC细胞相比,补肾活血方联合微囊化细胞移植技术能改善去卵巢大鼠激素分泌状态,其作为临床中医药预防和治疗卵巢早衰的一种手段具有良好的应景前景。
Objective: To investigate the effect of Bushen Recipe combined with microencapsulated cell transplantation on reproductive endocrine in rats. Methods: Fifty SD female rats were randomly divided into 5 groups: blank group (group A), ovariectomized group (group B), microencapsulated GC cell transplantation group (group C), low dose tonifying kidney medicine + microencapsulated GC Cell transplantation group (group D), high-dose kidney medicine + microencapsulated GC cell transplantation group (group E). Serum estradiol (E2), follicle stimulating hormone (FSH) and luteinizing hormone (LH) levels were measured by enzyme-linked immunosorbent assay (ELISA) Results: After transplanted with microencapsulated GCs, the level of serum E2 in group C was significantly higher than that in group B (P <0.05), and the levels of FSH and LH were significantly decreased (P <0.01) After GC cell transplantation, the levels of E2 in D and E groups were significantly increased compared with those in C group (P <0.05), and the levels of FSH and LH were significantly decreased (P <0.05). CONCLUSION: Compared with pure transplantation of microencapsulated GCs, Bushenhuoxue Decoction combined with microencapsulated cells can improve hormone secretion status of ovariectomized rats. As a means of clinical prevention and treatment of premature ovarian failure, TCM has a good Prospects.
